摘　　要：建立了微波水解-氨基酸自动分析仪测定酪蛋白胨中17种氨基酸含量的方法。样品经6 mol/L盐酸溶液150℃水解40 min后,通过氨基酸分析仪进行测定。结果表明酪蛋白胨中含有17种氨基酸,在相应浓度范围内,各氨基酸的峰面积与浓度线性关系良好,相关系数(r)均大于0.998。方法的最低检出限为0.02~1.22 g/kg,定量限为:0.07~4.05 g/kg,各氨基酸的回收率在80.6%~106.5%之间,RSD(n=6)均小于5.0%。该方法测定结果准确,可靠,可用于酪蛋白胨中氨基酸的测定。A microwave hydrolysis and amino acid automatic analyzer method was developed for determination of 17 amino acids in casein peptone.Aftere the samples were hydrolyzed by 6 mol/L hydrochloric acid solution at 150℃for 40min,and then determined by amino acid analyzer.The results showed that there were 17 amino acids in casein peptone,all the correlation coefficients(r)of 17 amino acids were greater than 0.998 in their linear ranges.The limits of detection were in the range of 0.02~1.22 g/kg,and the limits of quantification were 0.07~4.05 g/kg,the average recoveries of the samples were 80.6%~106.5%,and the relative standard deviation were less than 5.0%(n=6).The method is accurate and reliable,which is suitable for the determination of 17 amino acids in casein peptone.
