摘  要:目的研究DNA聚合酶ι(DNA polymerase iota,polι)基因对人类结肠癌化疗药物奥沙利铂敏感性的影响。方法采用RNAi技术使SW480细胞中polι基因的表达降低,通过Real time PCR实验检测基因polι低表达水平;运用MTT实验确定SW480细胞对药物奥沙利铂的敏感性确定给药剂量,并设立实验分组(空白组、对照组、实验组、空白加药组、对照加药组和实验加药组);分别使用流式细胞术和CCK-8实验分别检测不同组细胞的凋亡与增殖情况,通过集落形成实验检测细胞的存活数。结果RNA干扰技术成功降低了SW480细胞中polι基因的表达量(P<0.05),低表达polι基因并同时给予奥沙利铂药物的SW480细胞与只加药的对照组和空白组细胞相比,细胞凋亡实验显示细胞的凋亡率升高(P<0.01),CCK-8实验显示细胞的增殖率降低(P<0.05),集落形成实验显示细胞的存活分数降低(P<0.05)。结论下调SW480 polι基因表达水平,可提高人类结肠癌细胞对药物奥沙利铂的敏感性,并且可以抑制结肠癌细胞的增殖,促进其凋亡。Objective To investigate the effect of DNA polymerase iota(polι)gene on the sensitivity of the human colon cancer chemotherapeutic agent oxaliplatin.Methods Human colon cancer cell lines SW480 were transfected with polιgene-specific siRNA fragments.Real-time PCR was used to detect the interference efficiency of polιin the transfected cell lines.MTT assay was used to determine the sensitivity of SW480 cells to oxaliplatin and to determine the dose of oxaliplatin,and experimental groups(blank group,control group,experimental group,blank dose-adding group,control dose-adding group and experimental dose-adding group)were established.The apoptosis and proliferation of cells in different groups were detected by flow cytometry and CCK-8 assay respectively,and the survival number of cells was detected by colony formation assay.Results RNA interference successfully decreased the etpression of polιgene in SW480 cells(P<0.05).SW480 cells low expressing polιgene and administered with oxaliplatin showed increased apoptosis(P<0.01),decreased cell proliferation(P<0.05),and decreased cell survival number(P<0.05)compared to control and blank group cells with the addition of drug.Conclusion Down-regulation of SW480 polιgene expression levels significantly increased the sensitivity of human colon cancer cells to the drug oxaliplatin and inhibited the proliferation and promoted the apoptosis of colon cancer cells.
