乳腺癌组织中miR-216a-5p、TRIP4表达与患者病理特征和预后的关系

摘　　要：目的探讨乳腺癌组织中微小RNA(miRNA)-216a-5p、甲状腺激素受体相互作用蛋白4(TRIP4)表达与患者病理特征和预后的关系。方法选取85例乳腺癌患者,采用qRT-PCR法检测癌组织和癌旁组织中miR-216a-5p、TRIP4表达,并分析二者与患者临床病理特征的关系。随访3年,以二者均值将患者分为miR-216a-5p高表达组(≥0.430,44例)和miR-216a-5p低表达组(<0.430,41例),TRIP4高表达组(≥1.886,42例)和TRIP4低表达组(<1.886,43例),比较不同表达患者预后情况。结果乳腺癌、癌旁组织中miR-216a-5p相对表达量分别为0.430±0.109、1.012±0.166,TRIP4相对表达量分别为1.886±0.388、1.023±0.171,乳腺癌、癌旁组织中miR-216a-5p、TRIP4表达比较差异均有统计学意义(P均<0.05)。乳腺癌组织中miR-216a-5p与TRIP4表达呈负相关(r=-0.637,P<0.05),二者与人表皮生长因子受体-2(HER-2)表达、Ki-67增殖指数、TNM分期、淋巴结转移有关(P均<0.05)。miR-216a-5p高表达组3年无进展生存率、总生存率高于miR-216a-5p低表达组,而TRIP4高表达组3年无进展生存率、总生存率小于TRIP4低表达组(P均<0.05)。结论乳腺癌组织中miR-216a-5p低表达,TRIP4高表达,二者与HER-2表达、Ki-67增殖指数、TNM分期、淋巴结转移和预后有关。Objective To investigate the relationships between microRNA(miRNA)-216a-5p,thyroid hormone re⁃ceptor interacting protein 4(TRIP4)expression and the pathological parameters and prognosis of patients with breast can⁃cer.Methods Eighty-five cases of breast cancer patients were collected,and the expression of miR-216a-5p and TRIP4 in the cancerous tissues and adjacent tissues were detected by qRT-PCR.We analyzed the relationships between the two and the clinicopathological characteristics of patients.After 3 years of follow-up,they were divided into the high miR-216a-5p expression group(≥0.430,44 cases)and low miR-216a-5p expression group(<0.430,41 cases),high TRIP ex⁃pression group(≥1.886,42 cases)and low TRIP expression group(<1.886,43 cases).The prognosis of patients with dif⁃ferent expression was compared.Results The miR-216a-5p expression was lower in the breast cancer tissues(0.430±0.109)than that in the paraneoplastic tissues(1.012±0.166),and TRIP4 expression was higher(1.886±0.388)than that in the paracarcinoma tissues(1.023±0.171),with statistically significant difference(both P<0.05).The miR-216a-5p was negatively correlated with TRIP4 expression in breast cancer tissues(r=-0.637,P<0.05),and both were as⁃sociated with human epidermal growth factor receptor-2(HER-2)expression,Ki-67 proliferation index,TNM stage,and lymph node metastasis(all P<0.05).The 3-year progression-free survival rate and overall survival rate in the high miR-216a-5p expression group were higher than those in the low miR-216a-5p expression group,while the 3-year progression-free survival rate and overall survival rate in the high TRIP expression group were lower than those in the low TRIP expres⁃sion group(all P<0.05).Conclusion Low expression of miR-216a-5p and high expression of TRIP4 in breast cancer tissues are associated with HER-2 expression,Ki-67 proliferation index,TNM stage,lymph node metastasis and prognosis.
