摘  要:本文旨在实验教学中渗透各学科知识、融入学科发展的前沿知识,设计提取废弃菠萝皮中菠萝蛋白酶的实验。该实验通过简单的加热法制备低共熔溶剂(DES),并通过红外光谱以及粘度计等手段对制备的DES结构、物化性质进行表征,采用紫外分光光度计对提取的菠萝蛋白酶进行含量及活性测定,进一步积极引导同学们探究提取机理。该实验可以帮助学生掌握一些大型仪器的工作原理及操作技能,增强学生实验操作、谱图解析、提取机理探索等综合研究能力,树立废弃资源再利用的教学理念,实现科研和教学的相互促进。Aiming to infiltrate the knowledge of various disciplines and integrate the frontier knowledge of discipline development in experiment teaching,a comprehensive chemistry experiment was designed to extract bromelain from discarded pineapple peels.In this experiment,a deep eutectic solvent(DES)was prepared by a simple heating method.The structure and physicochemical property of the prepared DES was characterized by infrared spectroscopy and viscometer.The content and activity of the extracted bromelain were determined by ultraviolet spectrophotometer.Further actively guiding students to explore the extraction mechanism was also included.This experiment could help students master the working principles and operating skills of some large-scale instruments,and improve their research capabilities such as experimental operation,spectrogram analysis and extraction mechanism exploration.The process could also cause the student to establish a teaching concept of recycling waste resources,and achieve mutual promotion of scientific research and teaching.
