摘 要:目的:探讨食管鳞状细胞癌组织中叉头框转录因子M1(forkhead box transcription factor M1,FOXM1)和缺氧诱导因子-1α(hypoxia-inducible factor-1α,HIF-1α)的表达,并分析其与患者临床病理特征及预后的关系。方法:收集我院2017年06月至2018年07月收治的60例食管鳞状细胞癌患者的肿瘤组织及临床资料。采用免疫组织化学染色法检测FOXM1和HIF-1α蛋白在60例食管鳞状细胞癌组织、配对的癌旁正常组织中的表达;根据HIF-1α高/低表达、FOXM1高/低表达、HIF-1α及FOXM1共表达与否将其分为三种表达方式,分析FOXM1和HIF-1α表达与食管鳞状细胞癌患者临床病理特征及预后的关系。结果:FOXM1蛋白在食管鳞状细胞癌组织中的阳性表达率[68.33%(41/60)]显著高于远端正常食管组织[11.67%(7/60)](χ^(2)=40.139,P<0.001);HIF-1α蛋白在食管鳞状细胞癌组织中的阳性表达率[73.33%(44/60)]显著高于远端正常食管组织[6.67%(4/60)](χ^(2)=51.164,P<0.001);FOXM1的表达与HIF-1α的表达呈显著正相关(r=0.319,P=0.013)。FOXM1和HIF-1α阳性共表达与食管鳞状细胞癌的浸润深度(T期)有关(P<0.05)。FOXM1和HIF-1α阳性表达的食管鳞状细胞癌患者的总生存期(overall survival,OS)和无病生存期(disease-free survival,DFS)明显低于阴性表达者(P<0.05),且FOXM1和HIF-1α阳性共表达的食管鳞状细胞癌患者的生存期最短(P<0.05)。结论:FOXM1、HIF-1α的阳性表达与食管鳞状细胞癌密切相关,FOXM1和HIF-1α的联合检测可作为食管鳞状细胞癌患者预后预测的指标及潜在的治疗靶点。Objective:To explore the relationship between forkhead box transcription factor M1(FOXM1)and hypoxia-inducible factor-1α(HIF-1α)in esophageal squamous cell carcinoma tissues,and analyze their relationship with clinicopathological characteristics and prognosis.Methods:The tumor tissue and clinical data of 60 patients with esophageal squamous cell carcinoma admitted to our hospital from June 2017 to July 2018 were collected.Immunohistochemical staining method was used to detect the expression of FOXM1 and HIF-1αprotein in 60 cases of esophageal squamous cell carcinoma tissues and paired adjacent normal tissues.According to HIF-1αpositive/negative expression,FOXM1 positive/negative expression,HIF-1αand FOXM1 co-expression or not,etc.To divide them into three expression modes,and to analyze the relationship between the expression of FOXM1 and HIF-1αand the clinicopathological characteristics and prognosis of esophageal squamous cell carcinoma.Results:The positive expression rate of FOXM1 protein in esophageal squamous cell carcinoma tissues was 68.33%(41/60)significantly higher than 11.67%(7/60)(χ^(2)=40.139,P<0.001)in distal normal esophageal tissues.HIF-1αprotein positive expression rate in esophageal squamous cell carcinoma tissue was 73.33%(44/60)was significantly higher than that in the distal normal esophageal tissue 6.67%(4/60)(χ^(2)=51.164,P<0.001).FOXM1 expression and HIF-1αexpression was significantly positively correlated(r=0.319,P=0.013).FOXM1 and HIF-1αwere related to the depth of invasion(T stage)of esophageal squamous cell carcinoma(P<0.05).The overall survival(OS)and disease-free survival(DFS)of esophageal squamous cell carcinoma patients with positive expression of FOXM1 and HIF-1αwere significantly lower than those with negative expression(P<0.05),and patients with esophageal squamous cell carcinoma with positive co-expression of FOXM1 and HIF-1αhad the shortest survival(P<0.05).Conclusion:The positive expression of FOXM1 and HIF-1αis closely related to esophageal squamous cell carcino
