长链非编码RNA基因linc00152影响胃癌细胞周期和细胞迁移及侵袭的分子机制

摘　　要：目的探讨长链非编码RNA基因Linc00152促进胃癌细胞增殖、迁移和侵袭的分子机制。方法选取2022年1月至2022年2月间聊城市传染病医院收治的5例行胃癌切除术患者,采用qPCR法检测癌旁组织和胃癌组织以及胃癌细胞系中linc00152、miR-153-3p和FYN的相对表达量。采用shRNA和siRNA技术分别敲低linc00152和FYN的表达。采用流式细胞术检测shRNA敲低胃癌细胞SGC-7901中linc00152的表达后,细胞周期的变化。采用Transwell实验检测shRNA敲低胃癌细胞SGC-7901中linc00152的表达,以及siRNA敲低胃癌细胞SGC-7901中FYN的表达后,细胞迁移能力和增殖能力的变化。采用生物信息学分析linc00152和miR-153-3p的相互作用序列,并通过双荧光素酶报告基因检测验证该作用。结果长链非编码RNA Linc00152和FYN在胃癌组织和胃癌细胞中高表达,miR-153-3p在胃癌组织中低表达。Linc00152抑制miR-153-3p的表达,差异有统计学意义(P<0.05)。敲低linc00152和FYN后,胃癌细胞SGC-7901的细胞迁移能力和侵袭能力均显著降低,敲低linc00152还能将胃癌细胞SGC-7901和MKN-45的细胞周期阻滞在G0/G1期,差异有统计学意义(P<0.05)。结论在胃癌中linc00152是一种癌基因,linc00152的促癌作用轴为linc00152/miR-153-3p/FYN。Objective To explore the molecular mechanism of long non-coding RNA 152(linc00152)in promoting gastric cancer cell proliferation,migration and invasion.Methods qPCR was used to detect the relative expression of linc00152,miR-153-3 p and FYN in paracancerous tissues,gastric cancer tissues and gastric cancer cell lines.shRNA and siRNA were used to knock down the expression of linc00152 and FYN,respectively.Flow cytometry was used to detect the changes in cell cycle after shRNA knocked down the expression of linc00152 in gastric cancer cell SGC-7901.Transwell experiment was used to detect the changes in cell migration and invasion abilities after shRNA knockdown the expression of linc00152 in gastric cancer cell SGC-7901 and after knockdown of expression of FYN in gastric cancer cell SGC-7901 by siRNA.Using bioinformatics,the interaction of linc00152 and miR-153-3 p was analyzed,and the effect was verified by dual luciferase reporter gene assay.Results Long non-coding RNA linc00152 and FYN were highly expressed in gastric cancer tissues and gastric cancer cells,and miR-153-3 p was down-regulated in gastric cancer tissues.Linc00152 inhibits the expression of miR-153-3 p(P<0.05).After knocking down the expression of linc00152 and FYN,the cell migration and invasion ability of gastric cancer cells SGC-7901 were significantly reduced.Knockdown of linc00152 can also block the cell cycle of gastric cancer cells SGC-7901 and MKN-45 in G0/G1 phase.Conclusion Linc00152 is an oncogene in gastric cancer,and the action axis of linc00152 in promoting gastric cancer progression is linc00152/miR-153-3 p/FYN.
