摘 要:试验旨在研究新疆某区新引进腹泻症状妊娠母牛粪源产志贺毒素大肠杆菌(STEC)、‘the big six’血清学分布,为新引进妊娠母牛源STEC在肉牛产业链饲养阶段风险评估。随机采集新引进腹泻症状妊娠母牛的新鲜粪便,实验室分离、纯化培养,采用PCR方法检测STEC相关基因。结果显示,160株阳性菌株共分离出STEC编码毒力基因stx1 (13.13%)、stx2 (16.88%)、eae (23.13%)、hlyA (10.00%);阳性菌株共携带12种毒力基因谱,以编码毒力基因eae (11.25%)为主,其次为stx2+eae (7.50%)、hlyA (5.00%)、stx1+stx2 (3.75%)。共检出3种‘the big six’血清型,以O145检出率最高(4.38%),其次O157 (1.88%)、O121 (0.63%)。研究表明,引进妊娠母牛STEC相关毒力因子分离率较高,O145为优势血清型,需关注新引进母牛、新生犊牛、场内其他牛群消化道疾病症状,以防止外源致病菌的传播。The purpose of the experiment is to study the serological distribution of Shiga toxin producing Escherichia coli(STEC) and ’the big six’ from feces of newly introduced pregnant cows with diarrhea symptoms in Xinjiang,and to assess the risk of STEC from newly introduced pregnant cows in the feeding stage of beef cattle industry chain.Fresh feces of newly introduced pregnant cows with diarrhea symptoms were randomly collected,separated and purified in the laboratory,and the STEC related genes were detected by PCR.The results showed that the STEC encoding virulence genes stx1(13.13%),stx2(16.88%),eae(23.13%) and hlyA(10.00%) were isolated from 160 positive strains.The positive strains carried 12 kinds of virulence gene profiles,mainly encoding the virulence gene eae(11.25%),followed by stx2+eae(7.50%),hlyA(5.00%),stx1+stx2(3.75%).Three serotypes of ’the big six’ were detected,with the highest detection rate of O145(4.38%),followed by O157(1.88%) and O121(0.63%).The study indicates that the isolation rate of STEC related virulence factors of introduced pregnant cows is high,and O145 is the dominant serotype.Attention should be paid to the symptoms of digestive tract diseases of newly introduced cows,newborn calves,and other cattle on the farm to prevent the spread of exogenous pathogens.
