摘 要:目的 探究阿魏酸钠(SF)对氧诱导视网膜病变(OIR)小鼠肺组织损伤的影响。方法 建立OIR小鼠模型,用不同浓度的SF治疗。收集视网膜组织,通过血管闭塞面积和脉络膜厚度检测评估SF对视网膜新生血管(RNV)的影响;TUNEL法检测SF对视网膜细胞凋亡的影响;免疫组化检测SF对视网膜组织中血管生成相关蛋白HIF-1α和VEGF表达的影响。收集小鼠肺组织,通过肺组织的干湿(W/D)比值和病理学变化评估肺损伤;Western blot检测肺组织中HIF-1α、VEGF以及TLR4/NF-κB信号通路相关蛋白表达情况。结果 OIR模型小鼠视网膜组织中血管闭塞面积增大,视网膜厚度变薄,凋亡细胞增多,HIF-1α和VEGF表达上调(P<0.05)。另外,OIR模型小鼠肺组织W/D比值和肺损伤评分均增高;肺组织中HIF-1α、VEGF、TLR4、MyD88和NF-κB p65蛋白表达升高(P<0.05)。SF治疗可部分削弱OIR模型小鼠视网膜组织、肺组织的损伤,同时抑制TLR4/NF-κB通路激活(P<0.05)。结论 SF能够修复OIR小鼠RNV、抑制细胞凋亡、保护肺组织免受损伤,可能与抑制TLR4/NF-κB信号通路有关。Objective To investigate the influences of sodium ferulate(SF) on lung tissue damage in mice with oxygen-induced retinopathy(OIR).Methods The OIR mouse model was established and treated with different doses of SF.Retinal tissue was collected,and the influence of SF on retinal neo-vascularization(RNV) was evaluated by detecting vascular occlusion area and choroidal thickness;the TUNEL method was used to detect the effect of SF on retinal cell apoptosis;the influences of SF on the expressions of angiogenesis-related proteins HIF-1α and VEGF in retinal tissue were detected by immunohistochemistry.Mouse lung tissue was collected and lung damage was assessed by the wet/dry(W/D) ratio and pathological changes of lung tissue;the expressions of HIF-1α,VEGF and TLR4/NF-κB signaling pathway-related proteins in lung tissue were measured by Western blot.Results The occlusion area of blood vessels in retina tissue of OIR model mice increased,the thickness of retina became thinner and apoptotic cells increased,the expressions of HIF-1α and VEGF were up-regulated(P<0.05).In addition,the W/D ratio and lung damage score in lung tissue of OIR model mice were increased;the protein expressions of HIF-1α,VEGF,TLR4,MyD88 and NF-κB p65 were increased in lung tissue(P<0.05).SF treatment can partially alleviate the damage to the retina and lung tissue in OIR model mice,and inhibit TLR4/NF-κ Bactivation at the same time channel activation(P<0.05).Conclusion SF can repair RNV in OIR mice,inhibit apoptosis,and protect lung tissue from damage,which may be related to the inhibition of TLR4/NF-κB signaling pathway.
