摘 要:文章研究了川明参多糖超声波-微波协同提取的最佳工艺及体外抗氧化活性。通过单因素试验考察超声温度、超声时间、料液比和粉碎粒度对川明参多糖提取率的影响,确定各因素的最优水平。在单因素试验基础上,应用Box-Behnken试验设计和响应面分析法,探讨超声温度、超声时间、粉碎粒度对川明参多糖提取率的影响。响应面试验结果表明,超声波-微波协同提取川明参多糖的最优工艺为:超声温度52℃、超声时间31 min、粉碎粒度60目、料液比1:10(g/m L)。在此条件下,多糖提取率为25.12%,接近预测值,说明响应面优化的提取工艺参数可靠。体外抗氧化活性结果表明,川明参多糖具有一定抗氧化活性,对羟基自由基、DPPH自由基和超氧阴离子自由基具有一定清除能力,为川明参的高值化利用提供理论依据。The optimal technology and in vitro antioxidant activity of the polysaccharides from Chuanminshen Violaceum by ultrasonic-microwave synergistic extraction were studied.The effects of ultrasonic temperature,ultrasonic time,solid-liquid ratio and crushing mesh size on the extraction rate of C.violaceum polysaccharide were investigated by single factor experiment,and the optimal level of each factor was determined.On the basis of single factor experiment,the Box-Behnken experimental design and response surface analysis method were used to explore the effects of ultrasonic temperature,ultrasonic time and crushing mesh on the extraction rate of polysaccharide from C.violaceum.The results of the response surface methodology showed that the optimal process of ultrasonic-microwave synergistic extraction of C.violaceum polysaccharide was as follows:ultrasonic temperature 52℃,ultrasonic time 31 min,crushing mesh 60 mesh,and solid-liquid ratio 1:10(g/mL).Under the optimal process conditions,the extraction rate of polysaccharide was 25.12%,which was close to the predicted value,indicating that the extraction process parameters optimized by response surface methodology were reliable.The results of in vitro antioxidant activity showed that the polysaccharides of C.violaceum had certain antioxidant activity,and had a certain scavenging ability to hydroxyl radicals,DPPH free radicals and superoxide anion radicals,which provided a theoretical basis for the high-value utilization of C.violaceum.
