摘 要:目的研究组织蛋白酶(cathepsin G,Cat G)在脑出血(ICH)大鼠脑组织内的表达变化情况及其病理意义。方法选用成年雄性SD大鼠,通过向右侧基底核注射自体股动脉血建立大鼠ICH模型。本实验分为四组,分别是假手术组(Sham)、Sham加CatG抑制剂处理组(Sham+EGLINS),ICH组、ICH加Cat G抑制剂处理组(ICH+EGLINS),其中Sham组注射生理盐水。术后分别在第0.5天,1天,2天,3天,5天对各组大鼠进行神经功能损伤评分;蛋白印迹法检测Cat G,炎症因子IL-6,IL-1β,紧密连接蛋白(ZO-1)含量,干湿重法检测脑含水量。结果Cat G在ICH大鼠脑组织中显著上调,Cat G抑制剂EGLINS处理能显著抑制Cat G表达,并能抑制炎症因子表达,缓解因ICH诱导的局部脑水肿和血-脑脊液屏障(BCFB)破坏,改善大鼠的神经损伤。结论ICH损伤使大鼠脑组织中Cat G蛋白表达升高,而抑制Cat G表达能减轻炎症反应,缓解神经损伤,提示Cat G可能通过影响炎症反应参与了大鼠ICH诱导的神经损伤。Objective To explore the expression and pathological role of cathepsin G(Cat G)in brain of intracerebral hemorrhage(ICH)rats.Methods We randomly selected 80 adult male SD rats,collected autologous femoral artery blood and injected it into the right basal ganglia to make ICH model rats,and saline was injected in rats of the sham operation group.There are four groups in this study:sham group,sham+EGLINS group,ICH group,ICH+EGLINS group.Neurological deficit scores were evaluated at 0.5,1,2,3 and 5 days after surgery;The contents of Cat G、IL-6、IL-1β and ZO-1 were detected by Western blot,and the brain water content was detected by dry wet weight method.Results The expression of Cat G in brain t of ICH rats is significantly up-regulated.Cat G inhibitor eglins can inhibit the expression of cat g,reduce the expression of inflammatory factors,reduce brain water content,reduce the damage of(blood-cerebrospinal fluid barrier,BCFB)and reduce the degree of nerve injury.Conlusion The expression of Cat G protein in ICH rat brain increased,and the expression of Cat G inhibitor reduced the degree of nerve injury,suggesting that Cat G expression is related to inflammation and nerve injury in ICH rats.
