摘  要:将SD大鼠随机分为对照组、PTSD组、槲皮素(QUE)组,采用改良的连续单一应激(SPS&S)方法构建PTSD模型。造模后QUE组大鼠给予50 mg/kg QUE灌胃干预,分别在干预的第7、14 d做如下实验:旷场实验和高架十字迷宫实验测试大鼠行为表现;HE、Nissl染色法观察海马神经元结构变化;ELISA法测定海马组织IL-6含量;Western blot法检测海马组织BDNF蛋白水平;分析IL-6、BDNF水平与行为学改变之间的相关性。结果表明:大鼠PTSD样行为的发生与海马组织IL-6水平升高、BDNF水平降低呈显著相关,QUE对大鼠PTSD样行为有明显调节作用,其机制可能与降低海马组织IL-6含量,提高BDNF蛋白水平有关。SD rats were randomly divided into the QUE group,the control group and the PTSD group.The PTSD model was established by a combination of single continuous stress and plantar electric shock(SPS&S).The QUE group was treated with QUE by gavage(50 mg/kg)for 7 and 14 days respectively.Behavioral changes evaluat‐ed through open field and elevated plus maze.Structural changes of hippocampal neurons were observed by HE and Nissl staining.Western blot to measure BDNF protein,and Elisa method to detect interleukin-6(IL-6)in hippocampal tissue,the relationship between the expression level of IL-6 and BDNF in hippocampus and behavioral indexes was an‐alyzed by correlation analysis.Our results show that the occurrence of PTSD in rats is significantly related to an in‐creasing hippocampal IL6 level and a decreasing hippocampal BDNF level.QUE could improve the PTSD-like behav‐ior in rats by decreasing the level of hippocampal IL6 and increasing the content of hippocampal BDNF.
