摘 要:萝卜中含有的硫代葡萄糖苷(Glucoraphanin,GLR)经酶解作用会产生一种生物活性物质——莱菔素(Sulforaphene,SFE)。为了解不同品种萝卜种子中莱菔素的含量,选取三种不同品种的萝卜种子为研究对象,通过水解和用乙酸乙酯萃取的方法来提取莱菔素,并通过实验对液相色谱测定条件和莱菔素提取条件进行了优化,得到莱菔素的液相色谱检测条件为:反相C_(18)(250×4.6 mm,5μm),甲醇/水(30∶70,体积比),流速1.0 mL/min,进样量10μL,检测波长254 nm,柱温箱30℃。莱菔素的提取条件为反应温度25℃,固液比1∶40及反应时间2 h。As a biologically active substance,sulforaphene(SFE)is generated from glucoraphanin(GLR)contained in radishes by the enzymatic hydrolysis.In this study,three kinds of radish seeds were selected to explore the content of SFE in different kinds of radish seeds.SFE was extracted by hydrolysis and extraction with ethyl acetate.The determination conditions of high performance liquid chromatography(HPLC)and the extraction conditions of SFE were optimized.The HPLC conditions were as follows:reversed phase C_(18)(250×4.6 mm,5μm),methanol/water(30∶70,volume ratio),flow rate of 1.0 mL/min,injection volume of 10μL,detection wavelength of 254 nm,column temperature of 30℃.The extraction conditions of SFE were at 25℃,solid-liquid ratio of 1∶40 and reaction time for 2 h.
