摘 要:目的:对苗药苦皮藤药材正丁醇部位的薄层色谱条件进行提升优化。方法:择优筛选薄层板类型、展开系统、点样量、展距、显色剂,对苦皮藤药材正丁醇部位的儿茶素成分进行薄层色谱定性鉴别。结果:建立了以儿茶素为对照品,硅胶H板为展开板,点样量为2μL,苯:三氯甲烷:乙酸乙酯:正丁醇:甲酸(5:4:4:1:0.5)为展开剂,展距为8cm,显色剂为5%磷钼酸溶液的薄层色谱鉴定方法。试验得出贵州11批不同产地苦皮藤药材斑点清晰、分离度较好的薄层色谱图。结论:研究所建立的方法操作简便,专属性强、耐用性好、稳定性好,可用于苦皮藤药材正丁醇部位的鉴别。Objective To improve and optimize the thin layer chromatography conditions of n-butanol fraction of the seedling medicine Celastrus angulatus Maxim. Methods The type, unfolding system, sample size,spreading distance and chromogenic agent of the TLC were selected, and the catechin components in n-butanol part of the herb were identified by TLC. Results A TLC identification method was established with catechin as reference, silica gel H plate as unfolding plate, 2 μL sample size, benzene: trichloromethane: ethyl acetate:n-butanol: formic acid(5:4:4:1:0.5) as unfolding agent, spread distance of 8cm, and 5% phosphomolybdic acid solution as chromogenic agent. The thin layer chromatograms of 11 batches of Rhizoma fortunei from different producing areas in Guizhou were clear and well separated. Conclusion The method established in this study is simple, specific, durable and stable, and can be used for the identification of n-butanol parts of Celastrus angulatus Maxim.
