摘　　要：目的了解不同浓度Tagitinin D对人骨肉瘤MG63细胞增殖与凋亡的影响。方法将不同浓度Tagitinin D作用于MG63细胞24 h后,倒置显微镜观察细胞形态变化,MTS法检测Tagitinin D对MG63细胞增殖活性的影响,流式细胞术检测Tagitinin D对MG63细胞凋亡和周期的作用,细胞免疫荧光和Western-blot检测Tagitinin D对骨肉瘤细胞凋亡的影响。结果同一时间点,MG63细胞的生长抑制率随Tagitinin D浓度增加而升高(P<0.05或0.01)。随着Tagitinin D浓度升高,G0/G1期细胞比例下降,G2/M期和S期细胞比例升高,促凋亡蛋白Bax表达升高,抗凋亡蛋白Bcl-2表达降低(P<0.05或0.01)。结论Tagitinin D能够抑制MG63细胞增殖,阻滞细胞周期进展,促进MG63细胞凋亡。Objective To explore the effect of Tagitinin D of different concentrations on the proliferation and apoptosis of human osteosarcoma cell line MG63.Methods After MG63 cells were treated with Tagitinin D of different concentrations for 24 h,their morphological changes were observed with inverted microscope.The effect of Tagitinin D on the proliferation of MG63 cells was detected by MTS assay.The effect of Tagitinin D on the cell cycle and apoptosis of MG63 cells was detected with flow cytometry.The effect of Tagitinin D on the apoptosis of osteosarcoma cells was detected with immunofluorescence and Western-blot.Results At the same time point,the growth inhibition rate of MG63 cells increased with the increase of Tagitinin D concentration(P<0.05 or 0.01).With the increase of Tagitinin D concentration,the proportion of cells in G0/G1 phase decreased,the proportion of cells in G2/M phase and S phase increased,the expression of pro-apoptotic protein Bax increased,and the expression of anti-apoptotic protein Bcl-2 decreased(P<0.05 or 0.01).Conclusion Tagitinin D can inhibit proliferation,block cell cycle progression,and promote apoptosis of MG63 cells.
