摘 要:从湖南省长沙县养殖场牛胃秸秆发酵物中筛选获得了一株高产α-淀粉酶的菌株,命名为SCUEC8.通过对其形态学观察和生理生化特性,结合16S rDNA序列对比分析,初步鉴定该菌为枯草芽孢杆菌(Bacillus subtilis).探讨了不同碳源、氮源、培养条件等因素对枯草芽孢杆菌SCUEC8菌株产酶的影响,结果表明:在培养时间为16 h、pH为6.0、培养温度为37℃、碳源为20.0 g·L^(-1)的麦芽糖、氮源为15.0 g·L^(-1)的胰蛋白胨、金属离子为1.0 g·L^(-1)的Mn^(2+)时,枯草芽孢杆菌SCUEC8菌株生长量和α-淀粉酶的活性较高.A strain with high production ofα-amylase was selected from the straw fermentation of cattle stomach in Changsha County,Hunan Province,and was named SCUEC8.Based on the morphological observation,physiological and biochemical characteristics,and 16S rDNA sequence comparison analysis,the bacterium was preliminarily identified as Bacillus subtilis.Meanwhile,the effects of different factors such as carbon sources,nitrogen sources,and culture conditions on the enzymatic production of Bacillus subtilis SCUEC8 strain were investigated.The results demonstrated that the optimal culture conditions for the better growth of Bacillus subtilis SCUEC8 strain and higherα-amylase activity were as follows:16 h of culture time,pH 6.0,37℃of culture temperature,20.0 g·L^(-1) maltose as carbon source,15.0 g·L^(-1) tryptone as nitrogen source,and 1.0 g·L^(-1) Mn^(2+)as mental ion.
