摘  要:目的:探讨人胎盘脐带和绒毛来源的间充质干细胞(hPMSCs和UC-MSCs)对四氯化碳(CCL4)诱发的肝纤维化的改善效果。方法:根据处理方式不同将48只大鼠随机分为正常组、hPMSCs组、UC-MSCs组及肝纤维化模型组,每组各12只。通过腹腔注射CCL4构建大鼠肝纤维化模型,分别将hPMSCs和UC-MSCs通过尾静脉直接移植到大鼠体内;干细胞移植4周后,检测大鼠肝功能;取肝脏组织进行苏木精/伊红染色(HE)染色、马松(MTC)三色染色、天狼星红染色评估hPMSCs和UC-MSCs改善肝纤维化的效果;免疫组化法检测肝脏中转化生长因子-β1(TGF-β1)和α-平滑肌肌动蛋白(α-SMA)的表达水平。结果:与肝纤维化模型组比较,UC-MSCs和hPMSCs组均能够改善肝纤维化程度[(4.33±0.78)、(4.42±0.79)vs.(5.92±0.29)]和肝功能[白蛋白(ALB):(40.49±1.23)、(40.69±1.27)vs.(29.30±1.06)、丙氨酸氨基转移酶(ALT):(59.25±8.88)、(51.08±7.20)vs.(255.75±15.21)、碱性磷酸酶(ALP):(147.17±22.38)、(143.42±14.79)vs.(360.67±38.40)、天冬氨酸氨基转移酶(AST):(119.75±7.99)、(121.00±13.18)vs.(413.83±49.64)],组间差异具有统计学意义(P<0.05)。结论:脐带和绒毛来源的间充质干细胞的移植能够显著降低TGF-β1和α-SMA的表达量,改善CCL4诱发的肝纤维化。Objective:To evaluate improvement effect of mesenchymal stem cells(hPMSCsand UC-MSCs)from human umbilical cord and placental villi on carbon tetrachloride(CCL4)-induced rats of hepatic fibrosis.Methods:48 rats were randomly divided into normal group,hPMSCs group,UC MSCs group and liver fibrosis model group according to different treatment methods,with 12 rats in each group.Rat hepatic fibrosis model by CCL4-induced was established through intraperitoneal injection,and hPMSCs and UC-MSCs were transplanted into rats by tail vein.Liver function was detected after 4 weeks.Evaluation of the improvement effect of hPMSCs and UC-MSCs on liver fibrosis by HE staining,MTC trichromatic staining and Sirius red staining,and the expression levels of TGF-β1 andα-smooth muscle actin(α-SMA)were measured by immunohistochemistry.Results:hPMSCs and UC-MSCs could improve the degree of hepatic fibrosis(4.33±0.78,4.42±0.79 vs.5.92±0.29)and improve liver function in rats[ALB(40.49±1.23,40.69±1.27 vs.29.30±1.06),ALT(59.25±8.88,51.08±7.20 vs.255.75±15.21),ALP(147.17±22.38,143.42±14.79 vs.360.67±38.40),AST(119.75±7.99,121.00±13.18 vs.413.83±49.64)],and the differences were statistically significant(P<0.05).Conclusion:Mesenchymal stem cells from human umbilical cord and placental villus can significantly reduce the expression of TGF-β1 andα-SMA,and improve the liver fibrosis induced by CCL4.
