摘  要:目的 观察威廉姆斯综合征转录因子(WSTF)通过调控白细胞介素6(IL-6)/信号传导子及转录激活子3(STAT3)信号通路对乳腺癌MCF-7细胞增殖、侵袭及迁移能力的影响。方法 体外培养乳腺癌MCF-7细胞株,分为WSTF高表达组(转染慢病毒表达载体pLVX-PRDX4)、WSTF低表达组(转染慢病毒干扰载体pLVX-shPRDX4)、空载组(转染空载体pLVX-NC)和对照组(不作特殊处理)。各组转染48 h,采用CCK-8法检测细胞存活率,改良Matrigel Boyden小室实验检测侵袭细胞数,划痕实验检测划痕愈合率,流式细胞术检测细胞凋亡率,qRT-PCR法检测WSTF、IL-6和STAT3 mRNA表达,Western blotting法检测WSTF、IL-6和STAT3蛋白表达。结果 与对照组和空载组比较,WSTF低表达组细胞存活率、划痕愈合率和侵袭细胞数均降低,WSTF高表达组均升高(P均<0.05)。与对照组和空载组比较,WSTF低表达组细胞凋亡率升高,WSTF高表达组降低(P均<0.05)。与对照组和空载组比较,WSTF低表达组WSTF、IL-6和STAT3 mRNA及蛋白相对表达量均降低,WSTF高表达组均升高(P均<0.05)。结论 WSTF过表达可抑制乳腺癌MCF-7细胞凋亡,并促进其增殖、迁移和侵袭,其机制可能与激活IL-6/STAT3信号通路有关。Objective To observe the effects of Williams syndrome transcription factor(WSTF)on the proliferation,invasion,and migration of breast cancer MCF-7 cells by regulating the interleukin-6(IL-6)/signal transducer and activator of transcription 3(STAT3)signaling pathways.Methods Breast cancer MCF-7 cell lines were cultured in vitro and divided into high WSTF expression group(transfected with lentivirus expression vector p LVX-PRDX4),low WSTF expression group(transfected with lentivirus interference vector p LVX-sh PRDX4),empty load group(transfected with empty vector p LVX-NC),and control group(without special treatment).At 48 h after transfection,CCK-8 method was used to detect the cell survival rate,modified Matrix Boyden cell test was used to detect the number of invasive cells,Scratch test was used to detect the scratch healing rate,flow cytometry was used to detect the apoptosis rate,RT-q PCR was used to detect the expression of WSTF,IL-6 and STAT3 m RNA,and Western blotting was used to detect the expression of WSTF,IL-6 and STAT3 proteins.Results Compared with the control group and the empty load group,the cell survival rate,scratch healing rate and the number of invasive cells in the low WSTF expression group decreased,while those in the high WSTF expression group increased(all P<0.05).Compared with the control group and the empty load group,the apoptosis rate in the low WSTF expression group increased,while that in the high WSTF expression group decreased(both P<0.05).Compared with the control group and the empty load group,the relative m RNA and protein expression levels of WSTF,IL-6,and STAT3 in the low WSTF expression group decreased,while those in the high WSTF expression group increased(all P<0.05).Conclusion Overexpression of WSTF can inhibit the apoptosis of breast cancer cells and promote their proliferation,migration and invasion,which may be related to the activation of IL-6/STAT3 signaling pathway.
