摘　　要：目的系统评价新型抗癫痫药治疗癫痫的有效性和安全性。方法计算机检索PubMed、EMbase、The Cochrane Library、CNKI和WanFang Data数据库,搜集新型抗癫痫药卢非酰胺、唑尼沙胺、吡仑帕奈治疗癫痫的随机对照试验(RCT),检索时限均从2006年1月至2021年5月。由2名研究者独立筛选文献、提取资料并评价纳入研究的偏倚风险后,采用Rev Man 5.3软件和Stata 16.0软件进行Meta分析。结果共纳入16个RCT,包括4382例患者。Meta分析结果显示:新型抗癫痫药组治疗癫痫的有效率[RR=1.66,95%CI(1.45,1.89),P<0.00001]和完全控制率[RR=2.82,95%CI(2.01,3.96),P<0.00001]均优于对照组,且不增加严重不良反应发生率[RR=0.95,95%CI(0.72,1.27),P=0.75]。结论当前证据显示,新型抗癫痫药治疗效果良好,且具有良好的安全性。受纳入研究数量和质量的限制,上述结论尚待更多高质量研究予以验证。Objective To systematically review the efficacy and safety of new anti-epileptic drugs in the treatment of epilepsy.Methods PubMed,EMbase,The Cochrane Library,CNKI and WanFang Data databases were electronically searched to collect randomized controlled trials(RCTs)of new anti-epileptic drugs rufinamide,zonisamide,and perampanel in the treatment of epilepsy from January 2006 to May 2021.Two reviewers independently screened literature,extracted data and assessed the risk of bias of included studies;then,meta-analysis was performed by using RevMan 5.3and Stata 16.0 software.Results A total of 16 RCTs involving 4382 patients were included.The results of meta-analysis showed that the effective rate(RR=1.66,95%CI 1.45 to 1.89,P<0.00001)and seizure-free rate(RR=2.82,95%CI 2.01 to3.96,P<0.00001)in new anti-epileptic drugs group were higher than those in the control group,while it did not increase the serious adverse events(RR=0.95,95%CI 0.72 to 1.27,P=0.75).Conclusion Current evidence shows that new antiepileptic drugs have trends of better effectiveness,and their safety is satisfactory.Due to limited quality and quantity of the included studies,more high-quality studies are needed to verify above conclusion.
