摘 要:目的:探讨CD45^(+)红系祖细胞抑制进展期食管癌患者内皮祖细胞功能的作用及机制。方法:收集2018年5月1日至2019年5月31日来我院就诊的食管癌初治患者,将其分为早期肿瘤组和晚期肿瘤组,选取同时期来我院健康体检的志愿者作为对照组;流式细胞术检测外周血CD45^(+)红系祖细胞的比例、凋亡细胞的比例和细胞内ROS含量;Transwell小室进行细胞共培养实验;ELISA检测细胞培养上清中VEGF、SDF-1、MMP9和IL-10的含量。结果:对照组和早期肿瘤组外周血CD45^(+)红系祖细胞比例分别为1.39%和1.45%,差异没有显著的统计学意义;晚期肿瘤组患者CD45^(+)红系祖细胞比例为3.72%,相对于早期肿瘤组,差异明显;CD45^(+)红系祖细胞内ROS含量均显著高于CD45^(-)红系祖细胞,而各组CD45^(+)红系祖细胞内ROS含量均没有显著差异;晚期肿瘤组CD45^(+)红系祖细胞能够诱导内皮祖细胞的凋亡,抑制旁分泌VEGF、SDF-1、MMP9和IL-8。ROS抑制剂能够完全阻断CD45^(+)红系祖细胞的这些作用。结论:CD45^(+)红系祖细胞通过ROS通路,诱导进展期食管癌患者内皮祖细胞凋亡,并抑制其旁分泌功能。Objective:To investigate the inhibitory effect of CD45^(+)erythroid progenitor cells on the function of endothelial progenitor cells in patients with advanced esophageal cancer and its mechanism.Methods:The patients with primary esophageal cancer who came to our hospital from May 1,2018 to May 31,2019 were collected and divided into early cancer group and advanced cancer group.Volunteers who came to our hospital for health examination at the same time were selected as the control group.Flow cytometry was used to detect the proportion of CD45^(+)erythroid progenitor cells in peripheral blood,apoptotic cells and ROS content.Cell co-culture experiment was carried out in Transwell chamber.The contents of VEGF,SDF-1,MMP9 and IL-10 in cell culture supernatant were detected by ELISA.Results:The percentage of CD45^(+)erythroid progenitor cells in peripheral blood of control group and early cancer group was 1.39%and 1.45%,respectively,with no significant difference.The proportion of CD45^(+)erythroid progenitor cells in advanced cancer group was 3.72%,which was significantly different from that in early cancer group.The ROS content in CD45^(+)erythroid progenitor cells was significantly higher than that in CD45^(-)erythroid progenitor cells,but there was no significant difference in the ROS content in CD45^(+)erythroid progenitor cells among groups.CD45^(+)erythroid progenitor cells in advanced cancer group can induce the apoptosis of EPCs and inhibit the paracrine secretion of VEGF,SDF-1,MMP9 and IL-8.ROS inhibitors can completely block these effects of CD45^(+)erythroid progenitor cells.Conclusion:CD45^(+)erythroid progenitor cells can induce apoptosis and inhibit paracrine function of endothelial progenitor cells in patients with advanced esophageal cancer through ROS pathway.
