湖南省既往HIV-2抗体阳性血清RNA核酸检测结果分析

摘　　要：目的对湖南省既往人类免疫缺陷病毒-2(human immunodeficiency virus 2,HIV-2)抗体阳性者血清进行RNA核酸检测,以确定HIV-2感染。方法对2005—2019年湖南省HIV确证实验室保存的HIV-2抗体阳性血清样本进行核酸测序,采用自行设计的引物通过巢式PCR扩增HIV-2病毒核酸保守区gag基因片段(453 bp,HXB2620-1072),扩增产物测序并与HIV-2 gag基因参考序列比对判定。结果15份HIV-2抗体阳性血清样本中,13份(86.67%)HIV-2型核酸检测结果为阳性,2份未检出。13份HIV-2型核酸阳性样本中,gp36和gp105条带阳性率均为100.00%(13/13),gp41条带阳性率为92.30%(12/13),p51条带阳性率为69.23%(9/13),p31条带阳性率为84.62%(11/13)。结论通过核酸测序进一步验证湖南省已经存在HIV-2感染。Objective To analyze the RNA nucleic acid test results in serum samples with positive HIV-2 antibody to determine HIV-2 infection in Hunan province.Methods Fifteen confirmed HIV-2 antibody-positive serum samples collected in Hunan province from 2005 to 2019 were subjected to nucleic acid sequencing.The gag gene fragment(453 bp,HXB2620-1072),a conserved region of HIV-2 viral nucleic acid,was amplified by nested PCR using self-designed primers,and the amplified products were sequenced and compared with the reference sequence of HIV-2 gag gene.Results HIV-2 RNA was detected in 13(86.67%,13/15)samples and all(100.00%,13/13)showed both gp 36 and gp 105 bands,92.30%(12/13)had only gp 41 band.Conclusions The presence of HIV-2 infection in Hunan province has been further verified by nucleic acid sequencing.
