摘  要:钙调磷酸酶B样蛋白相互作用蛋白激酶(CIPK)是一种重要的Ca^(2+)离子信号传感器,在植物逆境胁迫应答和生长发育过程中发挥着重要作用。本研究利用同源比对方法从马铃薯全基因组中鉴定得到了27个StCIPK基因,并将其分为A~E 5个亚族。这些基因分布在马铃薯的12条染色体上,其编码蛋白长度为403~703 aa,等电点为5.03~8.20,分子质量为45 740.63~79 470.35 Da。该基因家族分为内含子缺失和内含子富集两类。亚细胞定位预测结果表明,StCIPK家族主要分布于细胞质和叶绿体。蛋白二级结构分析表明,StCIPK蛋白以α螺旋和不规则卷曲为主,β转角占比最少。StCIPK家族的启动子区域含有多个与抗逆境胁迫应答相关的应答作用元件。实时荧光定量PCR结果表明多数StCIPK基因能够强烈响应植物生长调节剂脱落酸处理。质量分数10%的聚乙二醇处理后,StCIPK09在24 h内均上调表达,StCIPK03在处理后6和12 h表达最高,分别是0 h的274.7和179倍。200 mmol·L^(-1)NaCl处理后6个基因相对表达量在2 h最高;StCIPK03在12 h响应强烈,为0 h的94.9倍。低温胁迫后6 h StCIPK03相对表达量为对照的286.4倍。本研究为马铃薯CIPK基因功能验证及利用分子育种进行抗逆遗传改良提供理论参考。Calcineurin B-like protein-interacting protein kinase(CIPK)is an important Ca^(2+) ion signal sensor,which plays an important role in plant stress response and growth and development.In this study,using the known Arabidopsis thaliana CIPK gene registration sequence,27 StCIPK genes were identified from the whole potato(Solanum tuberosum)genome by the method of alignment,and their bioinformatics and expression characteristics under simulated abiotic stress were analyzed.The results show that the potato StCIPK gene members can be divided into five subgroups(A–E),which are distributed on the 12 chromosomes of potato.The number of amino acids encoded by StCIPK gene ranges from 403 to 703 aa,the isoelectric point ranges from 5.03 to 8.20,and the molecular weight ranges from 45740.63 to 79470.35Da.Gene structure analysis showed that the gene family was divided into two categories,intron deletion and intron enrichment.The prediction of subcellular localization showed that the expression of StCIPK was mainly concentrated in the cytoplasm and chloroplast.Secondary structure analysis showed that StCIPK proteins are mainlyα-helices and irregular coils,whileβ-turns account for the least.The analysis of the cis-acting element results showed that the St CIPK family members contain multiple response elements related to cold resistance and resistance to adversity stress.The results of real-time fluorescence quantitative PCR showed that most of the potato CIPK genes can respond strongly to abscisic acid treatment.After10%polyethylene glycol treatment,the expression of St CIPK09 was up-regulated within 24 h,and the expression of St CIPK03 was the highest at 6 and 12 h after treatment,which were 274.7 and 179 times that of 0 h,respectively.The relative expression levels of the six genes were highest at 2 h after 200 mmol·L^(-1) Na Cl treatment,and St CIPK03 had a strong response at 12 h,which was 94.9 times that of 0 h.After the potato CIPK gene was treated with high temperature and low temperature stresses,the response of St C
