摘  要:目的研究大蒜素对诱导人卵巢癌细胞普通型HO-8910和高侵袭型HO-8910 PM凋亡的作用及机制,并对二者效果进行比较。方法将不同浓度的大蒜素作用于人卵巢癌HO-8910和HO-8910 PM细胞不同时间,进行PI/Hochest 33342荧光染色,采用逆转录聚合酶链反应(RT-PCR)和蛋白印迹(Western blotting)法检测survivin、Bcl-2、Bax基因和蛋白的表达。结果大蒜素能通过降低survivin基因和蛋白的表达增加Bax基因和蛋白的表达,通过降低Bcl-2基因的表达及Bcl-2/Bax比值明显促进HO-8910和HO-8910 PM细胞凋亡,且呈时间及剂量依赖性。大蒜素对HO-8910 PM细胞的作用均强于对HO-8910细胞的作用。结论大蒜素可通过调节survivin、Bcl-2家族蛋白诱导细胞凋亡,且对高转移性卵巢癌细胞更敏感。Objective To study the effect and mechanism of allicin on inducing apoptosis of common HO-8910 and highly invasive HO-8910 PM in human ovarian cancer cells.Methods Human ovarian cancer HO-8910 and HO-8910 PM cells were treated with different concentrations of allicin for different time,and PI/Hochest 33342 fluorescence staining was performed.The expressions of survivin,Bcl-2 and Bax genes and proteins were detected by reverse transcription polymerase chain reaction(RT-PCR)and Western blotting methods.Results Allicin could increase the expression of Bax gene and protein by reducing the expression of servivin gene and protein,and significantly promote the apoptosis of HO-8910 and HO-8910 PM cells by reducing the expression of Bcl-2 gene and Bcl-2/Bax ratio in a time-and dose-dependent manner.The effect of allicin on HO-8910 PM cells was stronger than that on HO-8910 cells.Conclusions Allicin can induce apoptosis by regulating survivin and Bcl-2 family proteins,and it is more sensitive to highly metastatic ovarian cancer cells.
