摘 要:目的探讨环状RNA SAMD4A(circSAMD4A)靶向微小RNA(miR)-410-3p对缺氧缺糖诱导的神经细胞损伤的影响。方法建立PC12细胞缺氧缺糖损伤模型。将PC12细胞分为对照(Con)组、模型(Model)组、Model+si-NC组、Model+si-circSAMD4A组、Model+miR-NC组、Model+miR-410-3p组、Model+si-circSAMD4A+anti-miR-410-3p组。实时定量PCR(RT-qPCR)检测circSAMD4A和miR-410-3p表达;四甲基偶氮唑蓝(MTT)法检测PC12细胞活力;流式细胞术检测PC12细胞凋亡率;商品试剂盒检测超氧化物歧化酶(SOD)活性、过氧化氢酶(CAT)活性、丙二醛(MDA)水平以及乳酸脱氢酶(LDH)释放量。双荧光素酶报告实验检测circSAMD4A和miR-410-3p靶向关系。结果与Con组比较,Model组PC12细胞的活力、SOD和CAT活性、miR-410-3p表达显著降低(P<0.05),凋亡率、MDA水平、LDH释放量、circSAMD4A表达显著升高(P<0.05)。与Model组、Model+si-NC组比较,Model+si-circSAMD4A组PC12细胞的活力、SOD和CAT活性、miR-410-3p表达显著升高(P<0.05),凋亡率、MDA水平、LDH释放量、circSAMD4A表达显著降低(P<0.05)。与Model组、Model+miR-NC组比较,Model+miR-410-3p组PC12细胞的活力、SOD和CAT活性、miR-410-3p表达显著升高(P<0.05),凋亡率、MDA水平、LDH释放量显著降低(P<0.05)。circSAMD4A和miR-410-3p直接结合。与Model+miR-410-3p组比较,Model+si-circSAMD4A+anti-miR-410-3p组PC12细胞的活力、SOD和CAT活性显著降低(P<0.05),凋亡率、MDA水平、LDH释放量显著升高(P<0.05)。结论干扰circSAMD4A通过靶向上调miR-410-3p表达可减轻缺糖缺氧诱导的神经细胞凋亡和氧化应激损伤。Objective To investigate the effects of cyclic RNA SAMD4A(circSAMD4A)targeting microRNA(miR)-410-3p on hypoxia and glucose deprivation-induced neuronal injury in vitro.Methods PC12 cells were cultured under hypoxia and glucose deprivation to establish a nerve cell injury model.PC12 cells were divided into control group,model group,si-NC group,si-circSAMD4A group,miR-NC group,miR-410-3p group,si-circSAMD4A+anti-miR-410-3p group.The expression levels of circSAMD4A and miR-410-3p were detected by Real-time quantitative PCR(RT-qPCR).Methyl thiazolyl tetrazolium(MTT)method was used to detect PC12 cell viability;flow cytometry was used to assess cell apoptosis;commercial kits were used to detect the superoxide dismutase(SOD)activity,and the levels of catalase(CAT)activity,malondialdehyde(MDA)and lactate dehydrogenase(LDH)release.moreover duble luciferase reporter experiment was used to detect the targeting correlation between circSAMD4A and miR-410-3p.Results Compared with those in control group,the cell viability,SOD and CAT activities,and miR-410-3p expression of PC12 cells in model group were significantly decreased(P<0.05),however,the apoptosis rate,MDA level,LDH release,and circSAMD4A expression were significantly increased(P<0.05).Compared with those in model group and si-NC group,the cell viability,SOD and CAT activity,miR-410-3p expression of PC12 cells in si-circSAMD4A group were significantly increased(P<0.05),but,the apoptosis rate,MDA level,and LDH release were significantly decreased(P<0.05).Compared with those in model group and miR-NC group,the cell viability,SOD and CAT activity,miR-410-3p expression of PC12 cells in miR-410-3p group were significantly increased(P<0.05),however,the apoptosis rate,MDA levels and LDH release were significantly decreased(P<0.05).Compared with those in miR-410-3p group,the cell viability,SOD and CAT activities of PC12 cells in si-circSAMD4A+anti-miR-410-3p group were significantly decreased(P<0.05),however,the apoptosis rate,MDA level,LDH release were increased significantl
