普鲁卡因调节lncRNA TTN-AS1表达对胃癌细胞恶性生物学行为的影响

摘　　要：目的探讨普鲁卡因调节lncRNA TTN-AS1表达对胃癌细胞增殖、侵袭和凋亡的影响。方法以人胃癌SGC-7901细胞为实验对象,采用MTT法、Transewell小室、Annexin V-FITC/PI双染法及qRT-PCR检测0.1 mmol/L、0.5 mmol/L和1.0 mmol/L的普鲁卡因对SGC-7901细胞增殖、侵袭、凋亡及TTN-AS1和miR-133b表达的影响。双荧光素酶报告基因实验及过表达或抑制TTN-AS1后miR-133b表达检测验证二者靶向关系,并进一步检测TTN-AS1靶向miR-133b对SGC-7901细胞增殖、侵袭和凋亡的影响。TTN-AS1 siRNA转染并结合普鲁卡因处理SGC-7901细胞,检测细胞增殖、侵袭和凋亡。结果不同浓度普鲁卡因可明显抑制SGC-7901细胞增殖和侵袭能力,促进细胞凋亡,下调TTN-AS1表达,上调miR-133b表达,呈现浓度依赖性(P<0.05)。抑制TTN-AS1表达可明显降低SGC-7901细胞增殖和侵袭能力,促进细胞凋亡,miR-133b inhibitor可明显减弱抑制TTN-AS1表达对细胞增殖、侵袭和凋亡的影响,普鲁卡因可明显增强抑制TTN-AS1表达对细胞增殖、侵袭和凋亡的影响。结论普鲁卡因可引起lncRNA TTN-AS1表达改变进而调控miR-133b表达,从而影响胃癌细胞增殖、侵袭和凋亡。Objective To investigate the effects of procaine on the malignant biological behavior of gastric cancer cells by regulating the expression of lnc RNA TTN-AS1 expression.Methods MTT assay,Transwell chamber assay,annexin V-FITC/PI double staining and qRT-PCR were used to detect the effects of 0.1mmol/L,0.5mmol/L and 1.0mmol/L procaine on the proliferation,invasion,apoptosis and expression levels of lncRNA TTN-AS1 and miR-133b in human gastric cancer SGC-7901 cells.Double luciferase reporter gene assay and miR-133b expression detection after overexpression or inhibition of TTN-AS1 were used to verify the targeting correlation between them.The effects of TTN-AS1 targeting miR-133b on proliferation,invasion and apoptosis of SGC-7901 cells were further detected.The proliferation,invasion and apoptosis of SGC-7901 cells were detected by TTN-AS1 siRNA transfection combined with procaine.Results The different doses of procaine could significantly inhibit the proliferation and invasion of SGC-7901 cells,promote cell apoptosis,down regulate the expression levels of TTN-AS11 and up regulate the expression levels of miR-133b in a concentration dependent manner(P<0.05).To inhibit the expression of TTN-AS1 expression could significantly decrease the proliferation and invasion of SGC-7901 cells and promote cell apoptosis.Moreover miR-133b inhibitor could significantly decrease the effects of TTN-AS1 inhibition on cell proliferation,invasion and apoptosis,and procaine could significantly enhance the effects of TTN-AS1 inhibition on cell proliferation,invasion and apoptosis.Conclusion The procaine can change the expression of TTN-AS1,so as to regulate the expression of miR-133b,thereby affecting the proliferation,invasion and apoptosis of gastric cancer cells.
