摘 要:目的检测上皮性卵巢癌组织中平滑肌蛋白22α(SM22α)基因启动子区甲基化状态及SM22α基因mRNA的表达情况,探讨其在卵巢癌发生发展中的作用。方法收集2015年12月至2019年10月住院手术治疗上皮性卵巢癌患者的卵巢癌组织标本51例为实验组,对照组收集同期55例因宫颈上皮内瘤变III级或宫颈癌行全子宫双附件切除的正常卵巢组织。应用甲基化特异性聚合酶链反应检测两组标本SM22α基因启动子区甲基化状态,qRT-PCR方法检测两组SM22αmRNA表达。结果卵巢癌组织中SM22α基因启动子区甲基化在卵巢癌组织中的阳性率显著高于正常卵巢组织(P<0.05),SM22α基因mRNA表达水平显著低于正常卵巢组织(P<0.05),且SM22α基因甲基化阳性的卵巢癌组织中SM22α的mRNA表达水平显著低于其在甲基化阴性组织中的表达(P<0.05)。此外,SM22α的甲基化状态及表达与卵巢癌患者淋巴结转移、FIGO分期有显著相关关系(P<0.05)。结论SM22α基因启动子区高甲基化状态导致SM22α表达下调可能促进卵巢癌的发生发展,SM22α有望成为卵巢癌诊断和治疗的分子靶点。Objective To investigate the methylation status and mRNA expression levels of SM22αin the tissues of ovarian cancer,and to explore its role in the occurrence and development of epithelial ovarian cancer.Methods A total of 51 ovarian cancer tissue specimens were collected in our hospital from December 2015 to October 2019,at the same time,55 normal ovarian tissue specimens were selected as control group.The methylation status of SM22αgene promoter was detected by using methylation-specific polymerase chain reaction,and the mRNA expression levels of SM22αwere detected by qRT-PCR.Results The methylation levels of SM22αpromoter region in ovarian cancer tissues were significantly higher than those in control group(P<0.05),however,SM22αmRNA expression levels were significantly lower than those in control group(P<0.05).Moreover,the SM22αmRNA levels in methylation positive ovarian cancer tissues were significantly lower than those in methylation negative ones(P<0.05).In addition,the methylation status and expression levels of SM22αwere significantly associated with FIGO stages and lymph node metastasis of patients with ovarian cancer(P<0.05).Conclusion The increased SM22αmRNA levels due to hypermethylation in its promoter region may be involved in the occurrence and development of ovarian cancer,which may be used as an important target for early detection and treatment of ovarian cancer.
