摘  要:目的通过分析大鼠颅盖骨生长发育过程中头颅各径线、颅骨厚度和脑容积的变化,以及局部黏着斑激酶(FAK)的表达规律,探寻颅骨快速生长期的标志物。方法实验采用SD大鼠,自出生后1~12周,测量其头颅各径线(A线为鼻尖至枕后粗隆的连线,B线为两侧眶内缘垂直于A线的连线,C线为两下颌角的连线,D线为两侧眶外缘的连线);将大鼠安乐死后剥离其两侧颅盖骨,分别使用量筒和游标卡尺测量其脑容积和颅骨厚度;应用实时定量PCR和蛋白质免疫印迹方法检测颅盖骨组织中FAK的mRNA和蛋白表达水平。结果(1)头颅径线:A线变化最为明显,第1~7周时的增长速度较快;第2~7周时,与前一周相比差异均有统计学意义(均P<0.05),而第7周后A线的增长趋于平缓。B线的增长较为平稳。第2周时C线和D线较上一周均明显增长(均P<0.05),第2周后两线的增长趋于平稳。(2)颅盖骨的厚度:第1~2周时颅盖骨几乎无增厚,但在第3~8周时较前一周增厚(均P<0.05),而且第8周后颅盖骨的厚度增长趋于平稳。(3)脑容积:1~3周的脑容积增长较快(P<0.001),第3周后趋于平稳。(4)脑容积与颅盖骨厚度的相关性分析结果:脑容积的增长与颅盖骨变厚呈正相关(R=0.90,P=0.001)。(5)FAK mRNA与蛋白表达变化情况:在大鼠脑发育过程中,FAK的mRNA和蛋白的表达量均逐渐升高,且均在第10周时达到最高值(均P<0.05),随后逐渐趋于平稳,蛋白与mRNA的表达趋势相一致。结论大鼠在出生后1~12周,FAK在颅骨组织中的表达趋势与颅骨的生长发育规律相一致。在脑完成快速生长后,FAK可能参与了大鼠颅骨的生长发育。Objective To investigate changes of skull diameters,skull thickness and brain volume and the expression of local focal adhesion kinase(FAK)during the growth and development of the calvaria of rats and to explore the markers of the rapid growth period of the skull.Methods SD rats were used in this study.Data were collected from 1 to 12 weeks after birth.The diameters of their heads were measured(Line A:Connecting the tip of the nose to the posterior occipital tuberosity;Line B:Connecting the two orbital margins perpendicular to line A;Line C:Connecting the two mandibular angles;Line D:Connecting the outer edges of the two orbits).After the rats were euthanized,the calvaria on both sides were peeled off,and the brain volume and skull thickness were measured with a graduated cylinder and a vernier caliper,respectively.The mRNA and protein expression levels of FAK in skull tissue were detected by real-time PCR and Western blot method.Results(1)Skull diameter line:Line A changed most obviously,and the growth rate was faster from 1 to 7 weeks,and from 2 to 7 weeks,the difference was statistically significant compared with that of the previous week(all P<0.05),and the growth flattened out after the seventh week.The growth of line B was relatively stable.In the second week,line C and line D both increased significantly compared with those of the previous week(both P<0.05),and the growth of the two lines became stable after the second week.(2)Thickness of calvaria:There was almost no thickening of calvaria at 1 to 2 weeks,but the thickness increased significantly at 3 to 8 weeks compared with those of the previous week(all P<0.05),and the increase in thickness became stable after 8 weeks.(3)Brain volume:The brain volume increased rapidly from 1 to 3 weeks(P<0.001),and became stable after the third week.(4)Correlation analysis results of brain volume and calvaria thickness:The increase of brain volume was positively correlated with calvaria thickening(R=0.90,P=0.001).(5)Changes of FAK mRNA and protein expression:During the
