摘 要:目的建立一种固相萃取-高效液相色谱法同时测定干虾制品(虾皮、海米、虾干)中7种人工合成着色剂(柠檬黄、苋菜红、胭脂红、日落黄、诱惑红、亮蓝、赤藓红)的方法。方法样品经提取液(无水乙醇∶氨水∶水=7∶2∶1,V/V/V)2次超声提取(提取液每次用量为15 ml),经Pro Elut PWA-2固相萃取柱净化后,以ZORBAX SB-C柱(250 mm×4.6 mm,5μm)为分离柱,0.02 mol/L乙酸铵和甲醇为流动相梯度洗脱,二极管阵列检测器在254 nm波长下检测,外标法定量。结果7种常见合成着色剂在15 min内完成分析,各合成着色剂在0.2μg/ml~20μg/ml浓度范围内与峰面积呈良好的线性关系,相关系数r>0.9999,检出限为0.01 mg/kg~0.03 mg/kg。实际样品检测中,合成着色剂的加标回收率为85.5%~97.2%,相对标准偏差(RSD)为2.13%~5.21%。结论该方法简单、快速、准确、高效,适用于干虾制品中合成着色剂的含量测定。Objective A method for the simultaneous determination of 7 kinds of synthetic colorants(Tartrazine,Amaranth,New Coccine,Sunset Yellow,Allura Red,Brilliant Blue and Erythrosine)in dried shrimp products was developed by solid phase extraction-high performance liquid chromatography(SPE-HPLC).Methods The samples were ultrasonically extracted twice by extracting solution(ethanol∶ammonia water∶water=7∶2∶1,V/V/V)(The volume of extracting solution was15 ml respectively),and then purified with Pro Elut PWA-2 SPE column.The extract was separated by ZORBAX SB-Ccolumn(250 mm×4.6 mm,5μm),and conducted for gradient elution with 0.02 mol/L ammonium acetate and methanol as mobile phases.The 7 kinds of synthetic colorants were detected by photo-diode array(PDA)detector at the wavelength of 254 nm and quantified by external standard method.Results Seven kinds of synthetic colorants could be separated in 15 min under the condition.The calibration curves showed good linearity within the concentrations of 0.2μg/ml-20μg/ml with the correlation coefficients(r)greater than 0.9999.The detection limits were 0.01 mg/kg-0.03 mg/kg.The recoveries of 7 kinds of synthetic colorants were 85.5%-97.2%,and the relative standard deviations(RSD)were 2.13%-5.21%.Conclusion The method is simple,rapid,accurate and efficient,which is suitable for the determination of 7 kinds of synthetic colorants in dried shrimp products.
