摘 要:目的检测糖尿病性白内障(DC)患者晶状体上皮细胞转录因子GATA结合蛋白4(GATA4)、糖原合酶激酶-3β(GSK-3β)信使核糖核酸(mRNA)表达水平,并分析二者与血脂、晶状体上皮细胞凋亡率的相关性。方法选取2018年3月—2021年12月余姚市人民医院收治的DC患者62例作为DC组,选取同时期60例无糖尿病的年龄相关性白内障患者作为对照组;检测所有患者血脂指标[总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)及三酰甘油(TG)]、晶状体上皮细胞GATA4、GSK-3βmRNA表达水平和凋亡率;Pearson法分析DC患者晶状体上皮细胞GATA4、GSK-3βmRNA表达水平的相关性,及二者分别与血脂指标、晶状体上皮细胞凋亡率的相关性。结果与对照组比较,DC组TC、LDL-C、TG、晶状体上皮细胞GATA4 mRNA表达水平、凋亡率升高,HDL-C、晶状体上皮细胞GSK-3βmRNA表达水平降低(P<0.05);DC患者晶状体上皮细胞GATA4 mRNA与GSK-3βmRNA表达水平成负相关(P<0.05);DC患者晶状体上皮细胞GATA4 mRNA表达水平与TC、LDL-C、TG、凋亡率成正相关,与HDL-C成负相关(P<0.05),GSK-3βmRNA表达水平与TC、LDL-C、TG、凋亡率成负相关,与HDL-C成正相关(P<0.05)。结论DC患者晶状体上皮细胞GATA4 mRNA高表达,GSK-3βmRNA低表达,二者与血脂水平及晶状体上皮细胞凋亡率密切相关,GATA4、GSK-3βmRNA可能是诊治DC的分子靶标,为治疗DC提供新思路。Objective This study aims to detect the levels of transcription factor GATA binding protein 4(GATA4)and glycogen synthase kinase-3β(GSK-3β)messenger ribonucleic acid(mRNA)in lens epithelial cells of patients with diabetic cataract(DC),and to analyze the correlation between the two factors and blood lipids and the apoptosis rate of lens epithelial cells.Methods A total of 62 cases of DC patients in Yuyao People’s Hospital from March 2018 to December 2021 were selected as the DC group,and 60 cases of age-related cataract patients without diabetes were selected as the control group;the blood lipid index[total cholesterol(TC),high density lipoprotein-cholesterol(HDL-C),low density lipoprotein-cholesterol(LDL-C)and triglycerides(TG)],the levels of GATA4 and GSK-3βmRNA levels and the apoptosis rate of lens epithelial cells in all patients were detected;Pearson method was used to analyze the correlation between the levels of GATA4 and GSK-3βmRNA in lens epithelial cells of DC patients,as well as their correlation between blood lipid index and apoptosis rate of lens epithelial cells.Results Compared with the control group,TC,LDL-C,TG,GATA4 mRNA levels and the apoptosis rate of lens epithelial cells in DC group increased,while the HDL-C and the level of GSK-3βmRNA in lens epithelial cells decreased(P<0.05);there was a negative correlation between GATA4 mRNA and GSK-3βmRNA expression in lens epithelial cells of DC patients(P<0.05);the level of GATA4 mRNA in lens epithelial cells of DC patients was positively correlated with TC,LDL-C,TG and apoptosis rate,and negatively correlated with HDL-C(P<0.05);the level of GSK-3βmRNA was negatively correlated with TC,LDL-C,TG and the apoptosis rate,and positively correlated with HDL-C(P<0.05).Conclusion The expression of GATA4 mRNA was high and GSK-3βmRNA was low in lens epithelial cells of DC patients.They were closely related to blood lipid level and apoptosis rate of lens epithelial cells.GATA4 and GSK-3βmRNA may be molecular targets for the diagnosis and treatment of DC
