摘 要:目的 基于简化基因组测序的基因分型技术(genotyping-by-sequencing, GBS)研究粗毛淫羊藿和巫山淫羊藿之间不同群体的差异和联系,为它们的起源和演化、育种研究提供分子生物学证据。方法 运用GBS技术对粗毛淫羊藿和巫山淫羊藿不同居群进行基因组测序,基于SNP标记对药材样品进行群体遗传学和全基因组关联研究,完成群体结构、群体历史和系统进化等分析。结果 共获得175.43G数据,测序Q30为94.92%,GC含量为42.92%,获得8 327 190个SNP。系统进化树与群体结构化分析、主成分分析结果具有一致性,贵州产巫山淫羊藿与重庆产的巫山淫羊藿存在一定的隔离,不同产地粗毛淫羊藿大部分聚类到一起,只有小部分与巫山淫羊藿更相近,同品种淫羊藿产地距离较近的群体也相对更相近。结论 GBS技术经济、高效、可靠,所获取的SNP数据量和数据质量可靠,建立的系统演化树、群体结构图、主成分分析聚类图可以相互补充、相互验证,对不同产地粗毛淫羊藿、巫山淫羊藿进行准确分类,可为淫羊藿植物基因分类和亲缘关系等遗传学研究提供可靠的技术支撑。Objective To study the differences and associations between Cumao Yinyanghuo(Epimedium acuminaum)and Wushan Yinyanghuo(Epimedium wushanense)based on Gnotyping-By-Sequencing and provide molecular biological evidence for their origin and evolution and breeding research.Methods The genomes of different populations of Cumao Yinyanghuo(Epimedium acuminaum)and Wushan Yinyanghuo(Epimedium wushanense)were sequenced by Gnotyping-By-Sequencing,and the population genetics and genome-wide association of the samples were studied based on SNP markers,and the population structure,population history and phylogeny were analyzed.Results A total of 175.43 G data was obtained,the Q30 was 94.92%,GC content was 42.92%,and 8,327,190 SNP were obtained.System evolutionary tree and group structured analysis,principal component analysis results had consistency.Wushan Yinyanghuo(Epimedium wushanense)production in Guizhou and Chongqing Wushan had isolation.Most Cumao Yinyanghuo(Epimedium acuminaum)from different habitats were clustered together,only a small percentage was the same as the Wushan Yinyanghuo(Epimedium wushanense).The populations with the same species of Epimedium with close habitats were also relatively close.Conclusion Gnotyping-By-Sequencing is economical,efficient and reliable,and the SNP data obtained was reliable in quantity and quality.The established phylogenetic tree,population structure diagram and principal component analysis cluster diagram can complement and verify each other,and accurately classify Cumao Yinyanghuo(Epimedium acuminaum)and Wushan Yinyanghuo(Epimedium wushanense)from different habitats.It can provide reliable technical support for genetic study of Epimedium plant gene classification and genetic relationship.
