摘 要:目的:探索补肾活血方对骨质疏松症模型大鼠骨密度的影响及其作用机制。方法:选取40只6个月龄雌性SD大鼠,运用随机数字表法将其分为正常组、模型组、中药治疗组和阳性对照组,每组10只。正常组仅切除卵巢周围脂肪组织,模型组、中药治疗组和阳性对照组切除双侧卵巢,饲养13周后,正常组及模型组予以蒸馏水灌胃,中药治疗组予以补肾活血方汤剂灌胃,阳性对照组予以利塞膦酸钠水溶液灌胃12周,测量大鼠脊柱骨密度后处死大鼠,分离大鼠双侧后肢股骨组织。Western Blot及免疫组化检测Notch1、Jagged1、Hes1蛋白表达水平。RT-PCR检测Notch1、Jagged1、Hes1 mRNA表达水平。结果:与正常组相比,模型组骨密度明显降低,差异有统计学意义(P<0.01)。与模型组相比,中药治疗组及阳性对照组骨密度明显升高,差异有统计学意义(P<0.01)。与正常组相比,Notch1、Jagged1蛋白表达量及基因表达量均明显下降,差异有统计学意义(P<0.01),Hes1蛋白表达量及基因表达量均明显升高,差异有统计学意义(P<0.01)。与模型组相比,中药治疗组及阳性对照组Notch1、Jagged1蛋白表达量及基因表达量均明显升高,差异有统计学意义(P<0.01),Hes1蛋白表达量及基因表达量均明显下降,差异有统计学意义(P<0.01)。从Western Blot及PCR检测结果可见,与阳性对照组相比,中药治疗组Notch1蛋白表达量更高,差异有统计学意义(P<0.05),Notch1基因表达量明显高于阳性对照组,差异有统计学意义(P<0.01)。Jagged1蛋白和基因表达量均明显低于阳性对照组,差异有统计学意义(P<0.01)。两组Hes1蛋白和基因表达量对比差异无统计学意义(P>0.05)。结论:补肾活血方能够改善骨质疏松症大鼠骨密度,其作用机制可能与Notch信号通路有关。Objective:To explore the effect of Bushen Huoxue recipe on bone mineral density and Notch signal pathway in model rats with osteoporosis.Methods:40 cases of 6-month-old female SD rats were randomly divided into normal group,model group,traditional Chinese medicine treatment group and positive control group with 10 rats in each group.The normal group only removed the adipose tissue around the ovary.The model group,the traditional Chinese medicine treatment group and the positive control group were removed both ovaries.After 13 weeks of feeding,the normal group and the model group were perfused with distilled water,the traditional Chinese medicine treatment group was perfused with Bushen Huoxue decoction,and the positive control group was perfused with sodium risedronate aqueous solution for 12 weeks.After measuring the bone mineral density of the spine,the rats were killed and the femoral tissues of both hind limbs were separated.The protein expression levels of Notch1,Jagged1 and Hes1 were detected by Western Blot and immunohistochemistry.The mRNA expression levels of Notch1,Jagged1 and Hes1 were detected by RT-PCR.Results:Compared with the normal group,the bone mineral density in the model group decreased significantly(P<0.01).Compared with the model group,the bone mineral density in the traditional Chinese medicine treatment group and the positive control group increased significantly(P<0.01).Compared with the normal group,the protein expression and gene expression of Notch1 and Jagged1 decreased significantly(P<0.01),and the protein expression and gene expression of Hes1 increased significantly(P<0.01).Compared with the model group,the protein expression and gene expression of Notch1 and Jagged1 in the traditional Chinese medicine treatment group and the positive control group were significantly increased(P<0.01),and the protein expression and gene expression of Hes1 were significantly decreased(P<0.01).Compared with the positive control group,the expression of Notch1 protein in the traditional Chinese medicin
