摘 要:[目的]考察蝉拟青霉5704s菌丝体提取液的降血糖作用,探索发挥降血糖作用的主要功能组分。[方法]制备蝉拟青霉5704s菌丝体提取液,分离提取液中峰1、峰2蛋白,采用小鼠试验和体外试验测定菌丝体提取液以及不同活性组分的降血糖作用,SDS-PAGE电泳测定主要降血糖组分分子量。[结果]蝉拟青霉5704s菌丝体提取液具有较好的降血糖作用,血糖下降率为36.85%;体外试验中,提取液峰1蛋白的α-糖苷酶抑制率为71.23%,峰2蛋白的抑制率为63.17%;小鼠试验中,提取液峰1蛋白的血糖下降率为40.23%,峰2蛋白的血糖下降率为25.70%;体外试验和小鼠试验均表明提取液峰1蛋白的降血糖效果优于峰2蛋白,峰1蛋白的分子量为57781 Da。[结论]确定蝉拟青霉5704s菌丝体提取液中发挥主要降血糖功效的组分是峰1蛋白。[Objective]To investigate the hypoglycemic effect of Paecilomyces cicadae 5704s mycelium extract,and to explore the functional components in the mycelium extract which play a main role in hypoglycemic effect.[Method]Prepared Paecilomyces cicadae 5704s mycelium extract,separated Peak 1 and Peak 2 proteins of the extract.Used mouse test and in vitro test to determine the hypoglycemic effect of mycelium extract and different active components.Then,sodium salti-polyacrylamide gel electrophoresis(SDS-PAGE)was used to determine the molecular weight of the main hypoglycemic components.[Result]The mouse test results showed that the Paecilomyces cicadae 5704s mycelium extract had a good effect on lowering blood sugar,and its blood sugar reduction rate was 36.85%.In the in vitro test,theα-glycosidase inhibition rate of the Peak 1 protein in the extract was 71.23%,and the Peak 2 protein was 63.17%.In the mouse test,the blood glucose drop rate of the Peak 1 protein was 40.23%,and the Peak 2 protein was 25.70%.Both the in vitro test and the mouse test all showd that the Peak 1 protein worked better than the Peak 2 protein in the blood sugar reduction effect.The molecular weight of the Peak 1 protein is 57781 Da as determined by SDS-PAGE.[Conclusion]It was determined that the main component in the extract of Paecilomyces cicadae 5704s mycelium that exerts the main hypoglycemic effect was the Peak 1 protein.
