摘 要:目的探究微小RNA(miR)-125b可否通过调节乳腺癌易感基因相关蛋白1(BAP1)调节胰腺癌细胞的发生与发展进程,探讨其在胰腺癌中的作用。方法采用实时定量聚合酶链反应和蛋白印迹检测2018年1月至2020年12月21例在我科接受手术患者的胰腺癌组织、癌旁组织和胰腺癌细胞系中miR-125b和其靶蛋白的表达水平。通过细胞克隆实验、流式细胞凋亡检测技术、划痕实验以及细胞侵袭实验检测转染前后胰腺癌细胞增殖、凋亡、细胞迁移和侵袭能力,组间比较采用t检验。结果miR-125b在胰腺癌组织中的表达显著上调(2.48±0.46比0.68±0.15,t=13.991,P<0.01)。过表达miR-125b组的BAP1的表达明显低于对照组(465.57±11.28比933.80±15.46,t=5.894,P<0.01)。过表达miR-125b组Panc-1和BxPC-3A细胞的增殖、迁移和侵袭能力明显高于对照组(214.56±8.45比78.43±5.15,t=3.482,P<0.01)。过表达BAP1组Panc-1和BxPC-3A细胞的增殖、迁移和侵袭能力明显低于对照组(30.85±4.48比220.63±16.17,t=3.956,P<0.01)。结论miR-125b可以通过靶向负调控BAP1的表达促进胰腺癌的进展。Objective To explore whether microRNA(miR)-125b can influence brca1 related protein 1(BAP1)to regulate the occurrence and progression of pancreatic cancer,and to investigate its functional effects on pancreatic cancer.Methods Real-time polymerase chain reaction and Western blotting were used to detect the expression level of miR-125b and its target protein in normal pancreatic tissue,pancreatic cancer tissue and pancreatic cancer cell line of 21 patients undergoing hepatobiliary and pancreatic laparoscopic surgery in our hospital from 2018.1 to 2020.12.Proliferation,apoptosis,migration and invasion of tumor cells were detected by cell cloning experiment,flow cytometry,scratch test and cell invasion test.T test was used for comparison between groups.Results The expression of miR-125b was significantly up-regulated in pancreatic cancer tissues(2.48±0.46 vs.0.68±0.15,t=13.991,P<0.01).The expression of BAP1 in miR-125b overexpression group was significantly lower than that in the control group(465.57±11.28 vs.933.80±15.46,t=5.894,P<0.01).The proliferation,migration and invasion abilities of PANC-1 and BXPC-3A cells in miR-125b overexpression group were significantly higher than those in the control group(214.56±8.45 vs.78.43±5.15,t=3.482,P<0.01).The proliferation,migration and invasion abilities of PANC-1 and BXPC-3A cells in BAP1 overexpression group were significantly lower than those in the control group(30.85±4.48 vs.220.63±16.17,t=3.956,P<0.01).Conclusion MiR-125b can promote the progression of pancreatic cancer by targeting negative regulation of BAP1 expression.
