结肠腺癌细胞微小RNA-21-5p和B细胞慢性淋巴细胞白血病/淋巴瘤7表达的关系及其临床意义

摘　　要：目的探讨微小RNA(miR)-21-5p/B细胞慢性淋巴细胞白血病/淋巴瘤7(BCL7A)轴在对结肠腺癌细胞系体外增殖和迁移的作用。方法通过生物信息学公共数据库GEPIA2和Starbase 2.0官网分析miR-21-5p和BCL7A在结肠腺癌中的表达特点,以及两者的相互关系。并开展体外细胞学研究,采用48孔板体外培养结肠腺癌细胞系Caco-2细胞,设立miR-21-5p模拟物组(miR-21-5p mimics)、mimics阴性对照组(mimics NC)、miR-21-5p抑制剂组(miR-21-5p inhibitor)、inhibitor阴性对照组(inhibitor NC)、miR-21-5p inhibitor+sh-BCL7A组,每组3个样本,均采用慢病毒转染实验过表达或敲减相应基因。基因干扰成功后,采用实时定量反转录聚合酶链反应(RT-qPCR)用于检测Caco-2细胞的BCL7A表达水平,噻唑蓝(MTT)法检测细胞增殖活力,划痕实验实验来评估细胞迁移率。结果生物信息学分析结果显示,结肠腺癌组织miR-21-5p表达水平为17.6±3.8,高于癌旁样本(11.5±2.6,t=7.587,P<0.05)。体外细胞学研究结果显示,miR-21-5p mimics组BCL7A表达水平为(98.5±10.5)%,高于mimics NC组[(44.5±5.2)%,t=7.982,P<0.05];miR-21-5p inhibitor组的细胞增殖活力(55.3±6.5)%,低于inhibitor NC组[(99.6±15.5)%,t=4.565,P<0.05];迁移率为(38.5±5.2)%,低于inhibitor NC组(75.6±12.0,t=4.913,P<0.05);miR-21-5p inhibitor+sh-BCL7A组的细胞增殖活力为(75.2±9.3)%,高于miR-21-5p inhibitor组[(55.3±6.5)%,t=3.038,P<0.05];迁移率为(55.3±8.2)%,高于miR-21-5p inhibitor组[(38.5±5.2)%,t=2.997,P<0.05]。结论miR-21-5p在结肠腺癌细胞增殖和迁移上具有重要作用。Objective To investigate the effects of microRNA(miR)-21-5p/B-cell CLL/lymphoma 7A(BCL7A)axis on the proliferation and migration of colon adenocarcinoma cell lines in vitro.Methods The expression of miR-21-5p and BCL7A and their correlation in colorectal adenocarcinoma were analyzed by bioinformatics public database GEPIA2 and Starbase 2.0 official website.An in vitro experiment was conducted in which the colon adenocarcinoma cell line CACO-2 cells were cultured in 48-well plates,and 5 groups were established:miR-21-5p mimics group,mimics NC group,miR-21-5p inhibitor group,inhibitor NC group,miR-21-5p inhibitor+sh-BCL7A group,with 3 samples in each.The corresponding genes were overexpressed or knocked down by a lentivirus transfection assay.After successful gene interference,real-time reverse transcription-polymerase chain reaction(RT-qPCR)was used to detect the expression level of BCL7A in CACO-2 cells,methyl thiazolyl tetrazolium(MTT)assay was used to detect the proliferation activity,and scratch test was used to evaluate the migration ability of Caco-2 cells.Results Bioinformatics analysis results showed that the expression level of miR-21-5p in colorectal adenocarcinoma tissue was 17.6±3.8,which was higher than that(11.5±2.6)in the adjacent samples(t=7.587,P<0.05).In vitro experiment results showed that the expression level of BCL7A in miR-21-5p mimics group was(98.5±10.5)%,which was higher than that(44.5±5.2)%in mimic NC group(t=7.982,P<0.05).The cell proliferation activity of miR-21-5p inhibitor group was(55.3±6.5)%,which was lower than that[(99.6±15.5)%]in inhibitor NC group(t=4.565,P<0.05);and the migration rate of miR-21-5p inhibitor group was(38.5±5.2)%,which was lower than that[(75.6±12.0)%]in inhibitor NC group too(t=4.913,P<0.05);The proliferation activity of miR-21-5p inhibitor+sh-BCL7A group was(75.2±9.3)%,which was higher than that[(55.3±6.5)%]in miR-21-5p inhibitor group,(t=3.038,P<0.05);and the migration rate of miR-21-5p inhibitor+sh-BCL7A group was(55.3±8.2)%,which was higher than t
