摘 要:目的探讨线粒体D-环区(D-Loop区)单核苷酸多态性(single nucleotide polymorphisms,SNPs)与喉癌发病风险的关系。方法实验标本取自河北医科大学第四医院耳鼻咽喉-头颈外科行喉癌手术的患者71例及在该院体检中心行健康查体的人群159例。采用聚合酶链反应(polymerase chain reaction,PCR)扩增目的片段,并进行线粒体D-Loop区测序。采用χ^(2)检验分析不同SNPs与喉癌发病风险之间的关系,并分析与喉癌发病相关的SNPs与患者临床特征的关系。结果线粒体DNA(mtDNA)D-Loop区的SNPs与喉癌的发病风险有关,73G、309C、315C及16319G基因型可能增加了喉癌的发病风险,而524C、556A、16288T、16291C及16311T基因型可能降低了喉癌的发病风险。结论线粒体D-Loop区SNPs可作为预测因子,用于提高喉鳞状细胞癌患者的早诊率,指导喉鳞状细胞癌患者的临床治疗。Objective To investigate the relationship between single-nucleotide polymorphisms(SNPs)in the mitochondrial DNA(mtDNA)D-Loop region and the risk of laryngeal carcinoma.Methods Experimental samples were collected from 71 laryngeal carcinoma patients who underwent surgery in the Department of Otolaryngology Head and Neck Surgery at the Fourth Hospital of Hebei Medical University.Samples were also collected from 159 healthy people who underwent physical examination in Medical Examination Center of our hospital.The target fragment was amplified by polymerase chain reaction(PCR),and the mtDNA D-Loop region was sequenced.Theχ^(2) test was used to analyze the relationship between different SNPs and the risk of laryngeal carcinoma,and the relationship between the SNPs associated with laryngeal carcinoma and the clinical features of these patients was also analyzed.Results SNPs in the mtDNA D-Loop region were related with the isk of laryngeal carcinoma.The genotypes of 73G,309C,315C and 16319G were significantly related with an increased risk in laryngeal carcinoma,whereas the genotypes of 524C,556A,16288T,16291C and 16311T were associated with the lower risk of laryngeal carcinoma.Conclusion SNPs in the mtDNA D-Loopregion can be used as predictors to improve the early diagnosis rate of laryngeal squamous cell carcinoma and guide the clinical treatment of the disease.
