摘 要:目的 观察罗汉松实多糖(PSP)对小鼠肝癌H22细胞移植瘤生长的影响,并探讨其可能机制。方法 取昆明小鼠54只,均在右腋皮下接种H22细胞建立移植瘤模型,造模成功后,将小鼠随机分为PSP高、中、低剂量组(PSP-H组、PSP-M组、PSP-L组)及环磷酰胺(CTX)组、香菇多糖(LNT)组、模型组(每组各9只),另取9只正常昆明小鼠作为对照组。PSP-H组、PSP-M组、PSP-L组分别给予480、240、120 mg/kg的PSP,CTX组给予20 mg/kg的CTX,LNT组给予100 mg/kg的LNT,对照组、模型组均给予10 mL/kg的生理盐水,各组均灌胃给药,1次/天,连续给药14 d。末次给药16 h后,摘眼球取血,处死小鼠,取肿瘤、胸腺和脾脏,称重,计算肿瘤抑制率及胸腺和脾脏脏器指数;HE染色法观察肿瘤组织病理变化;MTT法检测脾组织T、B淋巴细胞增殖能力;ELISA法检测眼球血清IL-2、IL-6、TNF-α、INF-γ。结果 与模型组比较,PSP-L组、PSP-M组、PSP-H组、CTX组、LNT组肿瘤质量降低(P均<0.05);与LNT组比较,PSP-M组、PSP-H组肿瘤质量降低(P均<0.05)。与LNT组比较,PSP-H组肿瘤抑制率升高(P<0.05)。与对照组比较,PSP-H组脾脏指数及PSP-L组、PSP-M组、PSP-H组、LNT组、模型组胸腺指数升高,CTX组、模型组脾脏指数低降低(P均<0.05);与模型组比较,PSP-L组、PSP-M组、PSP-H组、LNT组脾脏指数及胸腺指数升高,CTX组脾脏指数降低(P均<0.05);与LNT组比较,CTX组胸腺指数降低(P<0.05);与CTX组比较,PSP-L组、PSP-M组、PSPH组、LNT组脾脏指数和胸腺指数升高(P均<0.05)。模型组肿瘤细胞生长状态良好,清晰可见,数量多,排列紧密,细胞核深染,坏死细胞较少;CTX组肿瘤细胞数量减少,排列松散,出现不同程度的坏死区域;LNT组和PSP各组肿瘤组织疏松,肿瘤细胞减少,细胞不同程度坏死,其中PSP-H组细胞坏死更明显。与对照组比较,其余各组(除CTX组外)T、B淋巴细胞增殖能力增强(P均<0.05);与模型组比较,PSP-H组、LNT组T淋巴细胞增�Objective To observe the effect of polysaccharide from Podocarpus macrophyllus(PSP)on xenograft tu-mor growth of liver cancer H22 cells in mice,and to explore its possible mechanism.Methods H22 tumor cells weresubcutaneously inoculated into the right axilla of 54 Kunming mice to establish hepatocellular carcinoma transplanted tu-mor models. Then,tumor-bearing mice were randomly divided into the high-dose,medium-dose and low-dose PSP groups(PSP-H,PSP-M,and PSP-L groups),as well as cytoxan(CTX),lentinan(LNT),and model groups,with 9 mice in each group. Another 9 normal mice were selected as the control group. The PSP-H,PSP-M,and PSP-L groups were given480,240 and 120 mg/kg of PSP,respectively. CTX group was given 20 mg/kg of CTX,and LNT group was given100 mg/kg of LNT. The control and model groups were given 10 m L/kg normal saline. All groups were intragastrically ad-ministrated,once a day,for 14 consecutive days. At 16 h after the last administration,blood samples were collect-ed,mice were sacrificed,and tumor,thymus as well as spleen were isolated and weighted to calculate the tumor inhibitionrate and the organ indices. The pathological changes of tumor tissues were observed by HE staining and light microscope.MTT method was used to determine the proliferation abilities of T and B lymphocytes of spleen. Serum cytokines levels of in-terleukin-2(IL-2),IL-6,tumor necrosis factor-α(TNF-α)and interferon-γ(INF-γ)were measured by ELISA.Results Compared with the model group,the tumor weight of PSP-L,PSP-M,PSP-H,CTX and LNT groups decreased(all P<0. 05). Compared with the LNT group,the tumor weight of PSP-M and PSP-H groups decreased(both P<0. 05). Com-pared with the LNT group,the tumor inhibition rate increased in the PSP-H group(P<0. 05).
