半枝莲新克罗烷型二萜对人OCI-LY1淋巴瘤细胞增殖、凋亡及细胞周期的影响

摘　　要：目的研究细胞周期D类蛋白(cycnlin D3)在弥漫性大B细胞淋巴瘤OCI-LY1中的表达及半枝莲新克罗烷型二萜(半枝莲二萜J)对其的影响。方法CCK-8法检测细胞增殖活性,流式细胞仪检测细胞凋亡和细胞周期分布,Western blotting法检测cyclin D3蛋白表达。结果半枝莲二萜J对OCI-LY1细胞具有明显的增殖抑制作用;流式细胞仪检测显示,半枝莲二萜J诱导细胞凋亡呈剂量依赖性,且主要使细胞阻滞于G0/G1期,另外,G2/M期细胞也以剂量依赖的方式逐渐减少。Western blotting结果提示半枝莲二萜J以剂量依赖形式使cyclin D3蛋白表达降低。结论半枝莲二萜J具有抑制OCILY1细胞增殖并诱导其凋亡的作用,同时可使细胞阻滞于G0/G1期,通过下调cyclin D3表达发挥抗肿瘤作用。Objective To investigate the expression of cyclin D3 in human diffuse large B-cell lymphoma cell line OCI-LY1and the effects of a new neoclerodane diterpenoid from Scutellaria barbata(scutellone J,SJ)on it in vitro.Methods The effects of SJ on the growth of OCI-LY1 cells were detected by CCK8 assay.Apoptosis and cell cycle distribution were assessed by flow cytometer.The expression of cyclin D3 was checked by Western blotting.Results SJ markedly inhibited OCI-LY1 cell proliferation,as well as induction potency of apoptosis on OCI-LY1 cells in vitro in a dose-dependent manner by Annexin-V/PI double-labeled method.OCI-LY1 cells treated with SJ mainly showed accumulation in G0/G1 phase and reduction in the percentage of cells in G2/M phase in a dose-dependent manner.The cyclin D3 protein was decreased in OCI-LY1 cells treated with SJ.Conclusion SJ could inhibit the proliferation of human diffuse large B-cell lymphoma OCI-LY1 cells by regulating the cell cycle that arrests cells at G0/G1 phase.The antitumor role of SJ may be related to down-regulation of the expression of cyclin D3.
