摘 要:目的 基于生物信息学数据库分析上皮细胞转化序列2(ECT2)基因在乳腺癌中的表达及意义。方法 利用cBioPortal网站工具分析癌症基因组图谱(TCGA)数据库乳腺癌差异表达基因,人类蛋白质图谱(HPA)数据库分析ECT2蛋白在乳腺正常组织及乳腺癌组织中的表达,UALCAN分析CPTAC蛋白组学数据库和TCGA数据库中ECT2表达与临床病理参数的关系,GEPIA2分析TCGA数据库中ECT2表达与乳腺癌生存期的关系。结果 cBioPortal分析显示乳腺癌差异表达基因中高表达1 445个,选择前20个差异最显著的基因筛选出ECT2基因,其改变以扩增为主,获得次之,浸润性导管癌中扩增和获得频率最高;UALCAN分析基因热图同样显示ECT2在乳腺癌中过表达,其中在三阴性乳腺癌中表达较高;HPA数据库分析显示ECT2在正常组织及乳腺癌中均有表达,表达量居中;UALCAN分析显示,ECT2在浸润性导管癌中表达增高,在三阴性乳腺癌中呈高表达,并且与肿瘤分期、淋巴结转移、年龄分布及绝经期前后均有关(P<0.001);GEPIA2分析显示ECT2表达与乳腺癌总生存期及无病生存期均无相关性(P>0.05),但经过log-rank统计检验去除混杂因素后ECT2基因高表达患者总生存期及无病生存期均短于低表达患者。结论 基于多数据多组学高通量分析结果,推测ECT2在乳腺癌的发生发展过程中具有重要作用,可能是乳腺癌预后判断的有效指标及乳腺癌治疗的新靶点。Objective To analyze the expression and clinical significance of epithelial cell transforming sequence 2(ECT2) in breast cancer based on bioinformatics database. Methods CBioPortal was used to analyze the differentially expressed genes of breast cancer in TCGA database. Human protein atlas(HPA) database was used to analyze the expression of ECT2 protein in breast normal tissues and breast cancer tissues. UALCAN was used to analyze the relationship between ECT2 expression and clinicopathological parameters in CPTAC database and TCGA database, and GEPIA2 was used to analyze the relationship between ECT2 expression and survival of breast cancer in TCGA database. Results Among the 1 445 differentially expressed genes in breast cancer analyzed by c Bio Portal, ECT2 gene was screened from the top 20 genes with the most significant difference. Amplification was the main altered changes of ECT2 gene, followed by acquirement. And the frequency of amplification and acquirement was the highest in invasive ductal cancer. The gene heat map by UALCAN analysis also showed that ECT2 was overexpressed in breast cancer, especially in triple negative breast cancer. HPA database analysis showed that the expression level of ECT2 was moderate in normal tissues and breast cancer. UALCAN analysis showed that the expression of ECT2 was increased in invasive ductal carcinoma, and was high in triple negative breast cancer, it was related to tumor stage, lymph node metastasis, age distribution and menopause state(P<0.001). GEPIA2 analysis showed that there was no correlation between the expression of ECT2 and the overall survival or disease-free survival in TCGA database(P>0.05), however, after removing confounding factors by log-rank statistical test, the overall survival or diseasefree survival of patients with high expression of ECT2 gene was shorter than those with low ECT2 expression.Conclusion Based on the high-throughput analysis results of multi data and multi omics, it is speculated that ECT2 plays a very important role in the occu
