摘 要:毛霉病是一种具有血管侵袭性的感染性疾病,表现为局部血管浸润,血栓形成和组织坏死,阻碍病变部位白细胞渗出和抗真菌药物分布。葡萄糖调节蛋白78(glucose-regulated protein 78,GRP78)是调节毛霉侵袭血管的关键蛋白。在各类应激条件下内质网GRP78过表达并转运至细胞膜,成为毛霉进入血管内皮细胞的细胞表面受体。本综述回顾了GRP78介导毛霉感染的机制和病理生理学过程,总结了相关靶向药物的研究进展,为毛霉病的诊治提供参考。Mucormycosis is an infectious disease characterized by rapidly progressive vascular invasiveness,thrombosis and tissue necrosis,which could be potentially responsible for blocking the exudation of leukocytes to infection sites and affecting drug distribution.Glucose-regulated protein 78(GRP78)is a key protein involved in regulating the invasiveness of Mucorales.Endoplasmic reticulum GRP78 is overexpressed under various stress conditions and transported to the cell membrane to become a cell surface receptor for Mucorales entering into vascular endothelial cells.This article reviewed the mechanisms and pathogenesis of GRP78-mediated host cell invasion and summarized the progress in related targeted drugs,aiming to provide reference for developing multi-target intervention against mucormycosis.
