摘  要:目的优化岗梅根中总皂苷的提取工艺,并评价其中总皂苷抗氧化活性。方法以广西贵港野生岗梅根为原料,采用单因素试验及L_(9)(3^(4))正交试验优化岗梅根总皂苷回流提取工艺,以有机溶剂对粗提液进行萃取纯化,并以对1,1-二苯基-2-三硝基苯肼自由基(DPPH·)及·OH的清除效果评价岗梅根粗提液和纯化液的抗氧化活性。结果岗梅根中总皂苷最优提取工艺为70%乙醇,料液比1∶40(g∶mL),95℃温度下回流提取1.0 h,此工艺条件下总皂苷得率为5.99%。对DPPH·和·OH的清除能力由强到弱依次为萃取后水层>乙酸乙酯层>岗梅根粗提液,且岗梅根粗提液、萃取后水层、乙酸乙酯层中皂苷的质量浓度越大,清除DPPH·和·OH的效果越显著。结论优选工艺可为开发岗梅根总皂苷提供理论依据。岗梅根总皂苷具有抗氧化活性,有望作为天然抗氧化剂应用于食品、药品等领域。Objective To optimize the extraction process of total saponins from Ilicis Asprellae Radix,and to evaluate their antioxidant activity.Methods With the wild Ilicis Asprellae Radix from Guigang,Guangxi as the raw materials,the reflux-extraction process of total saponins from Ilicis Asprellae Radix was optimized by the single factor test and the L_(9)(3^(4))orthogonal test.The crude extract was extracted and purified with organic solvent,and the antioxidant activities of the crude extract and the purified solution of Ilicis Asprellae Radix were evaluated by the scavenging effects on 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH·)and·OH.Results The optimal extraction process of total saponins from Ilicis Asprellae Radix was as follows:the volume fraction of ethanol was 70%,the ratio of solid to liquid was 1∶40(g∶mL),and the total saponins from Ilicis Asprellae Radix were extracted in reflux at 95℃for 1 h.With this process,the yield of total saponins was 5.99%.The scavenging capacity to DPPH·and·OH was strongest in the water layer after extraction,followed by the ethyl acetate layer and the crude extract of Ilicis Asprellae Radix,and the scavenging effects of the crude extract of Ilicis Asprellae Radix,the water layer after extraction and the ethyl acetate layer on DPPH·and·OH increased with the increase of the mass concentration of saponins.Conclusion The optimal process can provide a theoretical basis for the development of total saponins from Ilicis Asprellae Radix.The total saponins from Ilicis Asprellae Radix have antioxidant activity,which are expected to be used in food,medicine and other fields as natural antioxidants.
