摘 要:目的:探讨Dickkopf相关蛋白3(DKK3)通过调控上皮细胞间质转化(EMT)相关蛋白表达对人前列腺癌PC3细胞凋亡、迁移和侵袭作用的影响。方法:选取2019年6月~2020年6月武汉大学人民医院确诊并进行前列腺癌根治手术的患者标本24例为研究对象,选择同期因良性前列腺增生行电切手术的患者标本24例为对照组,免疫组化法检测组织中DKK3表达情况。实时荧光定量PCR(RT-qPCR)法检测DKK3的mRNA表达水平,流式细胞术、MTT实验及Transwell实验测定过表达DKK3对前列腺癌细胞凋亡、增殖、迁移和侵袭的影响。Western blot法和qRT-PCR法测定过表达DKK3后EMT相关蛋白的表达变化。结果:前列腺癌组织较前列腺增生组织中DKK3表达明显降低。与正常前列腺上皮细胞系相比,前列腺癌PC3细胞中DKK3表达明显减少。与ctrl和NC组相比,ov-DKK3组细胞凋亡率显著升高,细胞增殖、细胞迁移和侵袭能力显著降低。ov-DKK3组中E-cadherin表达水平与ctrl和NC组相比表达升高,Vimentin和N-cadherin的表达水平较ctrl和NC组表达下降。结论:DKK3可以通过调控EMT相关蛋白的表达来影响前列腺癌PC3细胞的细胞凋亡及其迁移和侵袭能力。Objective:To investigate the effect on and mechanism of Dickkopf 3(DKK3)in the apoptosis,migration and invasion of human prostate cancer PC3 cells by targeting Epithelial-mesenchymal transition associated protein.Methods:Twenty-four patients with prostate cancer diagnosed and treated by radical prostatectomy in our hospital were included from June 2019 to June 2020 in the study group,and another 24 patients with benign prostatic hyperplasia undergone electrosurgical resection in the corresponding period were included in the control group.DKK3 expression in the tissues was detected by immunohistochemistry,and the effect of overexpression of DKK3 on apoptosis,proliferation,migration and invasion of prostate cancer cells were determined by flow cytometry,MTT assay and Transwell assay.The expression of EMT related proteins after overexpression of DKK3 were measured by Western blot and qRT PCR.Results:DKK3 expression was significantly lower in prostate cancer than in benign prostatic hyperplasia.Compared with normal prostate epithelial cell lines,the expression of DKK3 in prostate cancer PC3 cells was significantly down-regulated,yet the apoptosis rate in overexpression DKK3 group was significantly increased as compared with ctrl and NC group,and the ability of cell proliferation,migration and invasion was significantly declined.The expression level of E-cadherin in overexpression DKK3 group was higher than that in ctrl and NC groups,yet the expression levels of vimentin and N-cadherin were lower in ctrl and NC group.Conclusion:DKK3 can affect the apoptosis,migration and invasion of prostate cancer PC3 cells by regulating the expression of EMT related proteins.
