摘 要:目的研究miR-K1-5p相关的G1/S期基因编码的蛋白在卡波西肉瘤中表达的意义。方法利用生物信息软件miRDB和TargetScan Custom筛选miR-K1-5p的靶基因。使用生物信息数据库确定靶基因参与的信号通路,分析通路上的关键基因。应用免疫组化法检测靶基因及关键基因(miR-K1-5p相关的G1/S期基因)编码的蛋白在卡波西肉瘤和作为对照的血管瘤中的表达,并分析患者性别、年龄与蛋白表达以及各蛋白表达之间的相关性。结果细胞周期素依赖蛋白激酶抑制剂4(CDKN4)是miR-K1-5p的靶基因,参与细胞周期信号通路,通路下游关键基因为细胞周期蛋白A(CCNA)、细胞周期蛋白D2(CCND2)、周期素依赖性激酶2(CDK2)、周期素依赖性激酶4(CDK4)。免疫组化检测各基因编码的蛋白,CDKN4在卡波西肉瘤中的阳性率低于血管瘤,CCNA、CCND2、CDK2、CDK4在卡波西肉瘤中的阳性率高于血管瘤(P均<0.05);在卡波西肉瘤中CDKN4、CCNA、CCND2、CDK2和CDK4的表达与性别、年龄均无关(P均>0.05);在卡波西肉瘤中CDKN4与CCNA、CCND2、CDK2和CDK4的表达均呈负相关,CCNA、CCND2、CDK2、CDK4各蛋白之间的表达互为正相关(P均<0.05)。结论miR-K1-5p相关的G1/S期基因编码的蛋白(CDKN4、CCNA、CCND2、CDK2、CDK4)的异常表达在卡波西肉瘤发生、发展中可能发挥重要作用。Objective To study the expression significance of proteins encoded by miR-K1-5p related G1/S genes in Kaposi's sarcoma/KS.Methods Screening miR-K1-5p target genes through miRDB and TargetScan Custom of bioinformatics software,the bioinformatics database was used to identify the signal pathways involved by target genes and the key genes in the pathway was analyzed.Immunohistochemistry was used to detect the expression of proteins encoded by target gene and key genes(miR-K1-5p related G1/S genes)in Kaposi's sarcoma and hemangioma(the control group).The correlations between gender,age of patients and proteins expression and the correlations among proteins expression were analyzed.Results The target gene of miR-K1-5p was CDKN4,which was involved in the cell cycle pathway,and key genes were CCNA,CCND2,CDK2,CDK4 in the downstream of the pathway.The proteins encoded by genes were detected by immunohistochemistry.Positive expression rate of CDKN4 in KS was lower than that in the hemangioma group.Positive expression rates of CCNA,CCND2,CDK2 and CDK4 in KS were higher than the hemangioma group(P all<0.05).The expression of CDKN4,CCNA,CCND2,CDK2 and CDK4 in KS were not associated with gender and age(P all>0.05).CDKN4 was negatively correlated with the expression of CCNA,CCND2,CDK2 and CDK4 in KS.The expression of CCNA,CCND2,CDK2 and CDK4 were positively correlated in KS each other(P all<0.05).Conclusion The abnormal expression of proteins(CDKN4,CCNA,CCND2,CDK2,CDK4)encoded by miR-K1-5p related G1/S genes may play an important role in the occurrence and development of Kaposi's sarcoma.
