摘 要:目的探讨姜黄素对NF-κB通路和细胞周期蛋白D1(Cyclin D1)通路的调控作用,及对胰腺癌细胞增殖和凋亡的影响。方法通过浓度梯度姜黄素处理人胰腺癌细胞BxPC-3,采用CCK-8细胞活力法检测细胞活力并确定最适浓度,通过RT-PCR法检测Bax、Bcl-2、Cyclin D1 mRNA相对表达量,Western blot法检测Bax、Bcl-2、Cleaved Caspase3、p-NF-κB等蛋白的表达量。结果与对照组比较,不同浓度的姜黄素处理均使细胞活力明显降低,差异均有统计学意义(P<0.05),且呈剂量依赖性。Bcl-2、Cyclin D1的mRNA表达量明显降低,Bax的mRNA表达量明显升高,Bcl-2、Cyclin D1、p-NF-κB蛋白表达量明显降低,Bax、Cleaved Caspase 3蛋白表达量明显升高,差异均有统计学意义(均P<0.05)。结论姜黄素可诱导胰腺癌细胞凋亡,其作用机制与下调CyclinD1、抑制NF-κB活化有关。Objective To investigate the regulation of curcumin on the NF-κB pathway and Cyclin DI pathway,and its effect on the proliferation and apoptosis of pancreatic cancer cells.Methods A concentration gradient of curcumin was used to treat human pancreatic cancer cells BxPC-3,CCK-8 cell viability method was used to detect cell viability and determine the optimal concentration.RT-PCR was used to detect the relative expression of Bax,Bcl-2,and Cyclin DI mRNA.Western blot method was used to detect the expression of Bax,Bcl-2,Cleaved Caspase 3,p-NF-κB and other proteins.Results Compared with the control group,different concentrations of curcumin treatments significantly reduced cell viability,and the differences were statistically significant(all P<0.05)in a dose-dependent manner.The mRNA expression of Bcl-2 and Cyclin D1 was significantly reduced.The mRNA expression of Bax was significantly increased.The protein expression of Bcl-2,Cyclin DI,and p-NF-κB were significantly reduced.The protein expression of Bax and Cleaved Caspase 3 were significantly increased.The difference was statistically significant(all P<0.05).Conclusion Curcumin can induce apoptosis of pancreatic cancer cells,and its mechanism is related to down-regulation of Cyclin DI and inhibition of NF-κB activation.
