摘 要:为了筛选出生长性能和抗逆性良好的乳酸菌菌株,本试验用MRS培养基从蚯蚓肠道中分离得出3株候选乳酸菌,分别命名为R1、R2、R3,通过生化鉴定、16S rRNA测序、抑菌试验、药敏试验、耐酸试验、耐盐试验和动物安全性试验对其进行分析。结果显示:经生化鉴定和16S rRNA基因序列分析确认R1为植物乳杆菌,R2为乳酸片球菌,R3为肠膜明串珠菌。3株候选菌生长迅速、产酸能力强;对环丙沙星、庆大霉素和阿莫西林不敏感,对大肠埃希菌和金黄色葡萄球菌有较强抑制作用;且耐酸和耐胆盐。动物安全性试验未发现SPF级KM小鼠有异常和死亡现象。结果表明,本试验初步筛选出了3株生长性能优异、抗逆性好、具有优势潜力的乳酸菌菌株,为动物饲料添加剂的后续研究提供数据支持。In order to screen out lactic acid bacterial strains with excellent growth performance and high stress resistance, this study isolated three candidate strains(named R1, R2 and R3) of lactic acid bacteria from the intestines of earthworms using MRS medium, and further assessed by biochemical identification, 16 S rRNA sequencing analysis, bacteriostatic test, drug sensitivity test, acid resistance test, salt resistance test and animal safety test. The results of biochemical identification and 16 S rRNA gene sequence analysis showed that R1 was Lactobacillus plantarum, R2 was Pediococcus acidilactici, and R3 was Leuconostoc mesenteroides. These three candidate strains with rapid growth displayed strong acid-producing capacity, were insensitive to ciprofloxacin, gentamicin and amoxicillin, had strong inhibitory effect on Escherichia coli and Staphylococcus aureus, and were resistant to acid and bile salts. Animal safety test revealed no abnormality and death in SPF KM mice. Isolation of three strains of lactic acid bacteria with excellent growth performance, strong stress resistance and probiotic potential in this study thus builds a solid scientific basis for the follow-up research on animal feed additives.
