摘  要:优化气相色谱检测发酵液菌体中聚羟基脂肪酸酯含量的预处理过程。待测菌体经三氯甲烷及酯化液预处理,通过响应面优化分析方法建立二阶多项式,研究不同预处理参数对发酵液菌体中聚羟基脂肪酸酯检测含量的影响。预处理后的样品采用PH-5色谱柱(30 m×0.32 mm,0.25μm)检测。结果表明,当酯化温度为103℃,酯化时间为3.4 h,发酵液菌体样品质量浓度为12.64 g/L时,可以在较短的酯化时间内完成对菌体的预处理,保证了检测结果准确、可靠。To optimize the pretreatment process for determination of polyhydroxy acid esters in fermentation broth by gas chromatography. The bacteria to be tested were pretreated with trichloromethane and esterification solution, and the second-order polynomial was established by response surface optimization analysis method to study the effects of different pretreatment parameters on the detection content of polyhydroxy acid ester in the bacteria of fermentation solution. The pretreated samples were detected on a PH-5 column(30 m×0.32 mm, 0.25 μm). The results showed that when the esterification temperature was 103 ℃, the esterification time was 3.4 h, and the mass concentration of bacteria samples in fermentation broth was 12.64 g/L, the pretreatment of bacteria could be completed in a short esterification time, and the accuracy of detection could be ensured.
