摘  要:本文旨在探究AMPD1基因对静原鸡胸肌和腿肌肌肉组织中肌苷酸(IMP)的调控水平,为进一步探究静原鸡AMPD1基因的生物学功能奠定基础。本研究选择静原鸡高肌苷酸含量的胸肌和低肌苷酸含量的腿肌进行转录组测序分析,筛选出胸肌和腿肌组织差异表达的关键基因AMPD1,并进行GO注释和蛋白互作网络分析。利用RT-qPCR检测静原鸡肌肉组织中AMPD1基因的mRNA表达水平。从测序结果中筛选参与嘌呤代谢通路的关键基因AMPD1,注释结果显示AMPD1参与6个生物学过程,包括AMP脱氨酶活性、IMP的生物合成过程等。蛋白互作发现AMPD1与嘌呤核苷酸合成和代谢过程中的重要调控酶共享功能。实时荧光定量结果表明,静原鸡肌肉组织中AMPD1基因的表达量腿肌显著低于胸肌。综上所述,AMPD1基因可能是影响鸡肉IMP沉积的主效基因。研究结果可为进一步研究静原鸡IMP特异性沉积的AMPD1基因调控机制提供分子理论基础,同时为后续静原鸡AMPD1功能验证提供重要的科学依据和参考。The aim was to investigate the regulation level of AMPD1 gene on inosinic acid(IMP) in the muscle tissue of pectoral and leg muscles of Jingyuan chickens, and to lay the foundation for further investigation of the biological function of AMPD1 gene in Jingyuan chickens. In this study, the transcriptome sequencing analysis was carried out to identify the key gene, AMPD1, which was differentially expressed in pectoral and leg muscle tissues, and GO annotation and protein interaction network analysis were performed. The mRNA expression levels of AMPD1 gene in the muscle tissues of Jingyuan chickens were detected using RT-qPCR. The key gene AMPD1 involved in purine metabolic pathway was screened from the sequencing results, and the annotation results showed that AMPD1 was involved in six biological processes, including AMP deaminase activity and IMP biosynthesis process. Protein interactions revealed that AMPD1 shares functions with important regulatory enzymes in purine nucleotide synthesis and metabolism. The real-time fluorescence quantification results showed that the expression of AMPD1 gene in muscle tissue of Jingyuan chickens was higher in pectoral muscle than in leg muscle. Taken together, the AMPD1 gene may be the main effector gene affecting IMP deposition in chicken meat. The results can provide a molecular theoretical basis for further research on the regulatory mechanism of AMPD1 gene for IMP-specific deposition in jingyuan chickens, and also provide an important scientific basis and reference for subsequent validation of AMPD1 function in Jingyuan chickens.
