华蟾酥毒基对神经胶质瘤细胞增殖、迁移侵袭及凋亡的影响

摘  要:目的探讨华蟾酥毒基对人神经胶质瘤增殖、迁移侵袭及凋亡的影响及机制。方法以人胶质母细胞瘤U251作为主要研究对象,在不同浓度(0、0.25、0.5、1、2.5、5μmol/L)华蟾酥毒基作用24h后,采用MTT法检测其增殖能力;利用细胞克隆形成实验检测华蟾酥毒基对U251克隆形成能力的影响;通过伤口愈合实验和Transwell实验检测华蟾酥毒基对U251迁移和侵袭能力的影响;运用Western blot技术检测经华蟾酥毒基处理的U251细胞中Bax、Bcl-2蛋白表达水平的变化。结果华蟾酥毒基呈剂量依赖性抑制U251的增殖、迁移和侵袭;U251细胞中抗凋亡相关蛋白Bcl-2呈剂量依赖性下调趋势,促凋亡蛋白Bax表达呈剂量依赖性增高趋势。结论华蟾酥毒基能够明显抑制U251细胞的增殖、克隆形成及迁移侵袭能力,并通过调控Bax、Bcl-2蛋白的表达促进神经胶质瘤细胞的凋亡。Objective To investigate the effect and mechanism of cinobufagin on proliferation, migration, invasion and apoptosis of glioblastoma cells. Methods The effect of different concentrations of cinobufagin(0,0.25,0.5,1,2.5,5μmol/L) on the proliferation of U251 was detected by MTT method, and the effect of cinobufagin on the colony-forming ability of 251 was detected by cell clone formation assay.The effect of cinobufagin on the migration and invasion of U251 was detected by wound healing experiment and Transwell test, and the expression of Bax and Bcl-2 protein in U251 cells was detected by Western blot.Results Cinobufagin inhibited the proliferation, migration and invasion of glioblastoma U251 cells in a dose-dependent manner, decreased Bcl-2 expression, and elevated the expression of Bax.Conclusion Cinobufagin can significantly inhibit the proliferation, colony formation, migration and invasion of U251 cells, and promote the apoptosis of glioma U251 cells by regulating the expression of Bax and Bcl-2 proteins.
