摘 要:为探究氢供体Mg H2在GA合成受抑下对水稻种子萌发的影响,本试验在GA合成抑制剂烯效唑预处理12 h的基础上,分别加入不同浓度的氢供体Mg H2(0.005, 0.01, 0.02, 0.03, 0.04 mg/m L)培养水稻种子。记录水稻种子的萌发率及胚根胚芽的生长情况,测定萌发水稻种子总淀粉含量和H2O2水平,以及淀粉酶、α-淀粉酶相对活性、α-淀粉酶关键基因OsAmy 1A和OsAmy 3D的表达量。研究结果表明,在GA合成受抑下,一定浓度的氢供体通过提高H_(2)O_(2)的水平,上调α-淀粉酶基因OsAmy 1A和OsAmy 3D的表达,提高总淀粉酶和α-淀粉酶的活性,促进淀粉的降解,进而促进种子萌发及胚根和胚芽的生长,其中以浓度为0.02 mg/mL氢供体的作用最为明显。本研究结果为深入研究水稻种子萌发的机理提供了理论依据。To effect of hydrogen donor Mg H2on the germination of rice seeds when GA synthesis is inhibited. In this experiment, on the based of the pretreatment of 12 hours with GA synthesis inhibitor uniconazole, different concentrations of hydrogen donor Mg H2(0.005, 0.01, 0.02, 0.03, 0.04 mg/mL) were added to cultivate rice seeds.The germination rate of rice seeds and the growth of radicle and germ were recorded, and the total starch content and H2O2level of germinating rice seeds, as well as the relative activities of amylase, α-amylase and the expression of key α-amylase genes OsAmy 1A and OsAmy 3D were measured. Research indicates that under the inhibition of GA synthesis, hydrogen donor with a certain concentration up-regulated the expression of α-amylase genes OsAmy1A and OsAmy 3D by increasing the level of H_(2)O_(2), increased the activities of amylase and α-amylase, increased the degradation of starch, and promoted seed germination, radicle and germ growth, the effect of 0.02 mg/mL hydrogen donors is the most obvious. The results provide a theoretical basis for in-depth study of the mechanism of rice seed germination.
