摘 要:铁死亡(ferroptosis)是近年新发现的铁依赖的独特细胞程序化死亡方式,以致死性脂质过氧化蓄积为主要特征.目前,铁死亡研究领域发展迅猛,已成为生命科学及医学等领域的全球热点.大量研究提示铁死亡在心血管疾病、肝病、肾病、肿瘤及神经退行性等衰老相关重大疾病的发生发展及诊治中发挥重要作用.本文综述铁死亡的调控机制及其在脏器损伤和衰老相关疾病中的研究进展,总结现有铁死亡相关药物的应用,并对靶向铁死亡的临床转化应用前景进行展望.Ferroptosis is a recently discovered iron-dependent new form of programmed cell death,which is characterized by lethal accumulation of lipid peroxidation.This fast growing topic has become a global research hotspot in the fields of life science and medicine.Emerging evidence suggests ferroptosis plays an important role in the pathogenesis,diagnosis and treatment of various diseases such as cardiovascular disease,liver disease,kidney disease,tumor and aging-related neurodegeneration.This article reviews the current understanding of regulatory mechanisms of ferroptosis and its progress in organ injury and aging-associated diseases.We also summarize current potential applications of existing ferroptosis related drugs,and our perspectives with respect to future directions to target ferroptosis for clinical translational research.
