摘  要:目的 :研究脐带间充质干细胞来源外泌体(MSC-exo)对急性肺损伤大鼠模型的作用。方法 :利用超速离心法提取脐带间充质干细胞来源的外泌体,通过透射电镜、动态光散射及蛋白印迹实验鉴定外泌体。用脂多糖(LPS)诱导大鼠发生急性肺损伤动物模型,将动物分为阴性对照组、脂多糖(LPS)组和脂多糖+外泌体(LPS+exo)组。观察动物的精神状态并通过ELISA、QPCR、Western blot和组织化学分析等方法检测关键炎症因子的变化,分析外泌体对脂多糖(LPS)引起的急性肺损伤中炎症的调控作用。结果 :用脂多糖(LPS)成功诱导大鼠产生急性肺损伤动物模型,外泌体治疗可缓解急性肺损伤模型大鼠的体重减轻、缓解肺水肿、改善炎症因子蛋白水平、减少炎症细胞的浸润。结论 :脐带间充质干细胞来源外泌体能有效缓解脂多糖诱导的急性肺损伤表型及炎症水平。Objective To study the effect of umbilical cord mesenchymal stem cells derived exosomes(MSC-exo) on acute lung injury in rats. Methods The exosomes from umbilical cord mesenchymal stem cells were extracted by ultracentrifugation.The exosomes were identified by transmission electron microscopy, dynamic light scattering and Western blot. Lipopolysaccharide(LPS) was used to induce acute lung injury in rats. The rats were divided into several groups: negative control, lipopolysaccharide(LPS) and lipopolysaccharide + exosome(LPS+exo). The mental state of the animals was observed and the changes of key inflammatory factors were detected by ELISA, QPCR, western blot and histochemistry. The regulatory effect of exosomes on inflammation in lipopolysaccharide(LPS) induced acute lung injury was analyzed. Results The rat model of acute lung injury was successfully induced by lipopolysaccharide(LPS). Exosome therapy could alleviate the weight loss, pulmonary edema,protein levels of inflammatory factor and inflammatory cell infiltration in the rat model of acute lung injury. Conclusion The exosome derived from umbilical cord mesenchymal stem cells can effectively alleviate the phenotype and inflammatory level of acute lung injury induced by lipopolysaccharide.
