摘 要:目的 研究马钱苷元对白血病HL-60细胞生长及转化生长因子-β1(TGF-β1)表达的影响。方法 白血病HL-60细胞分为对照组(0μmol/L马钱苷元)、马钱苷元组(60、120、240μmol/L马钱苷元)、马钱苷元+PM组(240μmol/L马钱苷元和Hedgehog信号通路激活剂PM),CCK-8方法检测细胞增殖变化,PI单染法检测细胞周期变化,Annexin V-FITC/PI双染法检测细胞凋亡变化,Western blot方法检测细胞中p21、Bax、Bcl-2、TGF-β1、Smo、Gli2蛋白表达变化。结果 与对照组(0μmol/L)比较,60、120、240μmol/L的马钱苷元处理后白血病HL-60细胞OD值降低,G0/G1比例逐渐升高,细胞凋亡率逐渐升高,细胞中p21、Bax、TGF-β1蛋白表达水平逐渐升高,Bcl-2、Smo、Gli2蛋白表达水平逐渐降低,差异均有统计学意义(P<0.05)。与马钱苷元组比较,马钱苷元+PM组白血病HL-60细胞中Smo、Gli2蛋白水平升高,细胞OD值升高,细胞G0/G1比例降低,细胞凋亡率降低,细胞中p21、Bax、TGF-β1蛋白表达水平降低,Bcl-2蛋白表达水平升高,差异均有统计学意义(P<0.05)。结论 马钱苷元抑制白血病HL-60细胞生长,阻滞细胞周期,诱导细胞凋亡,促进细胞中TGF-β1蛋白表达,机制与抑制Hedgehog信号通路有关。Objective To investigate the effects of loganin on the growth of leukemia HL-60 cells and the expression of TGF-β1 in vitro.Methods Leukemia HL-60 cells were divided into control group(0μmol/L loganin), loganin group(60,120,240μmol/L loganetin), loganin+PM group(240μmol/L loganin and Hedgehog signaling pathway activator PM).The CCK-8 was used to detect cell proliferation;PI single staining was used to detect cell cycle;Annexin V-FITC/PI double staining was used to detect cell apoptosis;Western Blot was used to detect the expression levels of p21,Bax, Bcl-2,TGF-β1,Smo, and Gli2 protein.Results Compared with those in control group,the OD value in leukemia HL-60 cells was decreased after 60,120,240μmol/L loganin treatment, and the G0/G1 ratio and apoptosis rate as well as the expression levels of p21,Bax, and TGF-β1 proteins were gradually increased, however, the expression levels of Bcl-2,Smo, and Gli2 proteins were gradually decreased(P<0.05).Compared with those in loganetin group, the levels of Smo and Gli2 protein and OD value in loganetin+PM group were increased, but, the cell G0/G1 ratio and cell apoptosis rate as well as the expression level of p21,Bax and TGF-β1 protein were decreased, however, the expression levels of Bcl-2 protein were significantly increased(P<0.05).Conclusion The loganin can inhibit the growth of leukemia HL-60 cells, block the cell cycle, induce cell apoptosis, and reduce the expression levels of TGF-β1 protein in the cells, and itc action mechanism is related to the inhibition of Hedgehog signaling pathway.
